金复康口服液对人肺腺癌耐吉非替尼PC-9R细胞凋亡的影响

摘　　要：目的:探讨金复康口服液对人肺腺癌吉非替尼获得性耐药细胞株PC-9R的凋亡的影响及机制。方法:流式细胞术检测细胞周期及凋亡率,免疫组化法检测Caspase-3、Caspase-8的表达。结果:对于G1、G2期细胞数含量,金复康药物血清+吉非替尼组与金复康药物血清组、吉非替尼组比较,P<0.05,有统计学差异,对于S期细胞数含量,金复康药物血清+吉非替尼组、金复康药物血清组、吉非替尼组,P>0.05,无统计学差异,表明吉非替尼联合金复康药物血清后,金复康口服液增强了吉非替尼将细胞周期阻滞在G2期;在凋亡率方面,金复康药物血清+吉非替尼组与金复康药物血清组、吉非替尼组比较,P<0.05,有统计学差异,表明吉非替尼联合金复康药物血清后,金复康口服液增强了吉非替尼对PC-9R细胞的凋亡诱导作用;吉非替尼组、金复康药物血清+吉非替尼组与Con组比较,Caspase-3、Caspase-8的阳性表达率有统计学差异(P<0.05)。结论:金复康口服液能够逆转人肺腺癌吉非替尼获得性耐药细胞株对吉非替尼的凋亡抵抗,可能与上调caspase-3、caspase-8的表达有关。Objective ： To explore the influence of Jinfukang Oral Liquid on apoptosis and mechanism of human lung adenocarcina cell line PC -9R with Gefitinib resistant. Methods：Flow cytometry was used to detect the cell cycle and apoptosis rate and immunohistochemical method was used to detect the expressions of Caspase - 3 and Caspase - 8. Results ： G1 and G2 cycle cell number： compared Jinfikang Oral Liquid ＋ Gefutinib group with Jinfikang Oral Liquid group and Gefutinib group, there were statistical differences （P 〈 0.05）. S cycle cell number ： Jinfikang Oral Liquid ＋ Gefutinib group compared with Jinfikang Oral Liquid group and Gefitinib, it had no statistical differences（ P 〉 0.05 ）. It showed that Jinfukang Oral Liquid can enhance cell cycle block of Getifinib in G2. Apoptosis rate：Jinfikang Oral Liquid ＋ Gefitinib group compared with Jinfikang Oral Liquid group and Gefutinib group, there were statistical differences （P 〈 0.05 ）. It showed that Jinfukang Oral Liquid can enhance induction of apoptosis of Getifinib on PC - 9R. Positive expression rates of Caspase - 3 and Caspase - 8 ： Jinfikang Oral Liquid ＋ Gefitinib group compared with Jinfikang Oral Liquid group and Gefutinib group, it had statistical differences （ P 〈 0. 05 ）. Conclusion ： Jinfukang Oral Liquid can reverses resistance to apoptosis of human lung adenocarcina cell line PC -9R with Gefitinib resistant by increasing ex- pression rates of Caspase -3 and Caspase -8.
