摘 要:目的探讨TLR2和TLR4蛋白在肝癌组织中表达的意义,初步明确TLR2、TLR4与增殖相关因子在肝癌组织中表达的相关性。方法用SP免疫组化方法检测102例肝癌组织及癌旁组织、17例正常肝组织中TLR2和TLR4蛋白的表达,并结合临床病理参数进行分析。用Western-blot与Real-time RT-PCR方法检测60例新鲜冰冻肝癌组织及癌旁组织、17例正常肝组织中TLR2、TLR4、CyclinD1和p70s6k的蛋白表达及mRNA表达。应用SPSS13.0统计软件进行数据处理。结果 1肝癌组织中TLR2蛋白高表达33例(32.4%),低表达69例(67.6%);TLR4蛋白高表达42例(41.2%),低表达60例(58.8%)。TLR2和TLR4蛋白在肝癌组织中的表达显著高于癌旁组织和正常肝组织(P<0.05)。2TLR2蛋白的表达与肝癌的分化程度呈正相关(r=0.355,P<0.05)。TLR4蛋白的表达与肝癌的分化程度无关(r=0.096,P=0.338)。3Kaplan-Meier法生存分析显示TLR2蛋白的表达与术后无复发生存时间明显相关(Log rank:P<0.05)。4多因素Cox比例风险模型分析,结果TLR2蛋白的表达与预后明显相关(P<0.05)。5无论蛋白水平还是mRNA水平TLR2、TLR4的表达均与增殖相关因子CyclinD1和p70s6k的表达显著相关(r=0.715,P<0.05;r=0.713,P<0.05)。结论 TLR2和TLR4蛋白与肝癌的发生相关。TLR2与肝癌的分化和预后密切相关。TLR2和TLR4促进肝癌的增殖可能与CyclinD1和p70s6k有关。【Objective】To investigate the expression and significance of TLR2 and TLR4 in hepatocellular carcinoma.Preliminary to define the correlation of TLR2, TLR4 and proliferation-related factors in hepatocellular carcinoma.【Methods】S-P immunohistological assay was applied to detect the expression of TLR2, TLR4 in hepatocellular carcinoma tissues and adjacent normal tissues of 102 cases as well as normal liver tissues of 17 cases. Correlation analysis was used to determine the relation of TLR2, TLR4 with clinicopathological data. Western-blot and Real-time RT-PCR were applied to detect the expression of TLR2, TLR4, CyclinD1 and p70s6 k in 60 cases of hepatocellular carcinoma tissues, adjacent normal tissues and 17 cases of normal liver tissues. All statistical analysis were performed using the SPSS 13.0 version for Windows. 【Results】Among 102 cases of hepatocellular carcinoma,Higher expression of TLR2 was 33 cases(32.4%), lower expression was 69 cases(67.6%); Higher expression of TLR4 was 42 cases(41.2%), lower expression was 60 cases(58.8%). The expression of TLR2, TLR4 in hepatocellular carcinoma tissues was significantly higher than that in adjacent normal tissues and normal liver tissues(P〈0.05).The expression of TLR2 was positively correlated with the differentiation of hepatocellular(r =0.355, P〈0.05).The expression of TLR4 was independent of the differentiation of hepatocellular(r =0.096, P =0.338).The Kaplan-Meier survival analysis indicated that the expression of TLR2 was related to the postoperative relapse-free survival time(Log rank: P〈0.05). 4 Multivariate Cox proportional hazards model analysis pointed that the expression of TLR2 was related to prognosis(P〈0.05).The protein and mRNA levels of TLR2 and TLR4 were significantly related to CyclinD1 and p70s6k(r =0.715, P〈0.05; r =0.713, P〈0.05).【Conclusions】TLR2 and TLR4 proteins were related to the initiation of hepatocellular.
