摘　　要：目的研究乳腺增生症、乳腺导管内癌和浸润性乳腺癌组织中mdm2蛋白的表达及siRNA mdm2对肿瘤细胞增殖的影响。方法采用免疫组织化学方法检测乳腺增生症、乳腺导管内癌和浸润性乳腺癌组织中mdm2蛋白的表达;将siRNA mdm2质粒转染乳腺癌MCF-7细胞,应用MTT检测siRNA mdm2对肿瘤细胞增殖的抑制作用。结果 mdm2蛋白在乳腺导管内癌组织和浸润性乳腺癌组织中的阳性表达率均为100%(18/18,20/20),在乳腺增生症组织中表达阳性率为20%(4/20)。与乳腺增生症组织比较,乳腺导管内癌组织和浸润性乳腺癌组织中mdm2蛋白阳性表达率明显增高(P<0.01)。MTT结果表明转染pGCsiRNA-mdm2后MCF-7细胞生长受到抑制,与mock组和pGCsiRNA-scramble组比较差异有统计学意义(P<0.01)。转染pGCsiRNA-mdm2 72 h后与48 h比较,MCF-7细胞生长受抑制明显,差异有统计学意义(P<0.01)。结论 mdm2在乳腺癌组织中存在广泛表达,其阳性表达可能促进肿瘤细胞的增殖过程。Objective To explore the expression of mdm2 in human cyclomastopathy, ductal carcinoma in situ and invasive breast cancer tissues, and to study the effects of mdm2 siRNA on cell proliferation in MCF-7 cells. Methods The expression of mdm2 was studied by means of immuno- histochemistry in cyclomastopathy, ductal carcinoma in situ and invasive breast cancer tissues. MCF-7 cells were transfected with pGCsiRNA-mdnr2 and pGCsiRNA-scramble by Lipofectamine 2000. Cell proliferation was measured by MTY assay. Results The results of immunohistochemistry in- dicated that the positive rates of mdm2 was 100% in ductal carcinoma in situ tissues and invasive breast cancer tissues, which was only 20% in cyclo- mastopathy tissues. Compared with cyclomastopathy tissues, the positive rates of mdm2 protein was obvious increased in ductal carcinoma in situ tis- sues and invasive breast cancer tissues （P 〈 0.01 ）. Compared with mock and pGCsiRNA-scramble, pGCsiRNA-mdm2 significantly inhibited the survival rates in MCF-7 cells （P〈 0.01 ）. Conclusion Mdm2 generally expressed in breast cancer tissue and promote the process of tumor prolifer- ation.
