摘 要:目的探讨心理应激对大鼠颞下颌关节髁突软骨细胞基质金属蛋白酶-3(MMP-3)和金属蛋白酶组织抑制剂-3(TIMP-3)表达的影响及其机制。方法将48只成年Wistar雄性大鼠随机分为实验3w组、实验6w组、实验去除刺激6w组(恢复组)和空白对照组,每组12只。对3个实验组大鼠施加电击、制动、气候箱、食物诱惑等刺激,使大鼠处于心理应激状态。将各实验组大鼠分别于3w、6w和刺激去除后6w处死;对照组同恢复组大鼠一起处死。取颞下颌关节标本制作切片,免疫组织化学染色,光镜下观察髁突软骨细胞MMP-3和TIMP-3的表达情况。结果 4组大鼠颞下颌关节髁突软骨细胞中MMP-3表达强度不同,差异有统计学意义(P<0.05);实验3w组和实验6w组MMP-3的表达强度均高于对照组(P<0.05)。4组大鼠颞下颌关节髁突软骨细胞中TIMP-3表达强度不同,差异有统计学意义(P<0.05);恢复组TIMP-3的表达明显高于对照组(P<0.05)。结论 MMP-3和TIMP-3分别在实验3w、6w组和恢复组高表达,MMP-3和TIMP-3表达平衡可能是应激影响髁突软骨的途径之一,合理调控该平衡可能会阻断应激对软骨的破坏。Objective To investigate the mechanism and influence of Matrix Metalloproteinase-3 (MMP-3) and Tissue Inhibitor of Metalloproteinase-3 (TIMP-3)expression in the temporomandibular joint condylar cartilage of rat under psychological stress.Methods 48 adult male Wistar rats were randomly divided into the experimental group 3 w,experimental group 6 w,experimental group 6 w removal stimulation (recov-ery group)and control group,with 12 rats in each group.The rats in 3 experimental groups were subjected to the following stimulating factors such as electroshock,keeping stillness,climate change and food tempting.Making them under psychological stress.The rats in control group were not subjected.Then the experimental group rats were executed at three weeks,six weeks and six weeks after ceasing stimulation, each time being twelve rats.The control group rats were executed at the same time as the recovery group. Take temporomandibular joint specimens to produce slices,immunohistochemical staining,observe theexpression of MMP-3 and TIMP-3 in condylar cartilage cells under light microscopy.Results The MMP-3 expression intensity of 4 groups in the temporomandibular joint condylar cartilage cell were intensity dif-ferent (P〈0.05);Experiment 3 weeks and 6 weeks of MMP-3 expression were significantly greater than the control group (P〈0.05).The TIMP-3 expression intensity of 4 groups in the temporomandibular joint condylar cartilage cell were different too(P&lt;0.05);The expression of TIMP-3 in recovery group was sig-nificantly greater than the control group (P〈0.05).Conclusion In the experimental group and the recov-ery group,MMP-3 and TIMP-3 respectively have high expression as cartilage cell's iconic cytokines of carti-lage cell damage and repair.MMP-3 and TIMP-3′s expression balance may be one way of stress affecting condylar cartilage.Reasonable regulation of the balance may be block the destruction of the cartilage stress.
