摘  要:目的:探讨神经节苷脂GM3在多囊卵巢综合征大鼠卵巢发育过程中的表达及定位,为进一步研究神经节苷脂GM3在卵泡发育和排卵过程中的作用提供依据。方法:采用高性能薄层色谱分析法测定PCOS大鼠卵巢神经节苷脂GM3的表达;用免疫荧光激光共聚焦技术检测神经节苷脂GM3在PCOS大鼠的不同发育阶段卵泡中的表达及定位。结果:多囊卵巢综合征大鼠卵巢组织中提取的神经苷脂主要成分为GM3;与正常对照组相比,PCOS大鼠卵巢组织中神经苷脂GM3的表达水平显著降低。PCOS大鼠的初级卵泡、次级卵泡及窦状卵泡中的神经苷脂GM3的表达水平显著降低。结论:多囊卵巢综合征大鼠的卵巢发育过程中神经苷脂GM3的表达水平显著降低。提示,卵泡发育异常可能与GM3的表达程度密切相关。[ Abstract] Objective: To explore the expression and location of ganglioside GM3 during ovarian development of rats with polycys- tic ovary syndrome (PCOS), provide a basis for further researching the effect of ganglioside GM3 during ovarian development and ovulation. Methods : High - performance thin - layer chromatogram was used to detect the expression of ganglioside GM3 in ovary of PCOS rats. Immu- nofluoreseence combined with laser confoeal microscopy was used to detect the expression and localization of ganglioside GM3 in follicles in different developmental stages of PCOS rats. Results: The key component of ganglioside abstracted from ovarian tissue of PCOS..rats was GM3 ; compared with normal control group, the expression level of ganglioside GM3 in ovarian tissue of PCOS rats decreased significantly. The expression levels of ganglioside GM3 in primary follicles, secondary follicles and antral follicles of PCOS rats decreased significantly. Conclusion: The expression level of ganglioside GM3 during ovarian development of PCOS rats decreases significantly, which indicates that abnormal ovarian development may be closely correlated with expression level of ganglioside GM3.
