摘  要:目的以梅花鹿鹿角盘为原料,运用响应曲面法优化鹿角盘胶原蛋白提取工艺;方法以胶原蛋白提取率为指标,在单因素的基础上,选取pH值、加酶量、酶解温度、酶解时间4个因素进行4因素4水平的BoxBehnken试验设计,采用响应曲面法分析4个因素对胶原蛋白提取率的影响;结果鹿角盘胶原蛋白的最佳提取工艺,根据实际情况修正为:pH1.8、加酶量4%、酶解温度37℃、酶解时间4 h,在此条件下,鹿角盘胶原蛋白提取率可达到80.10%。结论鹿角盘胶原蛋白的提取受很多因素的影响,因此,对多种因素进行单因素和交互影响分析,优化实验方法及条件,会大大提高鹿角盘胶原蛋白的提取率。Objective With the antler base of Cervus Nippon,extracting technology of collagen was optimized by the response surface methodology.Methods Based on single factor experiments,a four-factor and four-level Box-Behnken design was carried out to investigate the effects of pH value,enzyme loading,hydrolysis temperature,hydrolysis time with collagen extraction ratio as index.Effects of the 4 factors on collagen extraction ratio were analyzed by the method of response surface.Results According to the actual situation,the optimal extracting technology of collagen of antler base of Cervus Nippon was revised as follows:under the condition of pH1.8,enzyme loading 4%,hydrolysis temperature 37 ℃ and hydrolysis time 4 h,collagen extraction ratio of antler base of Cervus Nippon could reach 80.10%.Conclusion It was influenced by many elements for the extraction of collagen of antler base of Cervus Nippon.Therefore,analyzing the effects of single factors and interaction of each other and optimizing experimental methods and conditions would greatly improve the extraction ratio of collagen of antler base of Cervus Nippon.
