摘  要:目的为了提高志贺氏菌的分离率,选用硫酸丁胺卡那霉素作为抑菌剂。方法将添加与不添加硫酸丁胺卡那霉素的肉汤,按AOAC验证体系进行比对,并增加耐药以及不同形式的染菌试验。结果添加硫酸丁胺卡那霉素可以使志贺氏菌增菌肉汤增菌液BAX-PCR的Ct值下降1~5个值。对测试方法和参考方法进行卡方检验,χ2都小于3.84(P〉0.05),说明两方法没有显著性差异。结论当Ct值≤25.5时,传统方法可完全分离出目的菌。相同Ct值条件下,XLD和HE平板的分离效果要高于R&F和MAC。R&F可用于可疑菌落的初步鉴定。测试方法确定的阳性样本数大于参考方法,测试方法优于参考方法。Objective Amikacin Sulphate is selected as a bacteriostatic agent in order to raise the isolation rate of Shigella from Shigella broth (SB). Methods The enrichment broth in the presence and absence of Amikacin Sulphate are compared on the basis of AOAC validation system. Simultaneously, Shigella - resistance and various spiked tests are performed. Results The BAX - PCR test results show that Amikacin Sulphate can drop the Ct value of the SB broth by 1 - 5. An examination of test method and reference method by chi - square ( x2 ) all show x2 〈 3.84 (P 〉 O. 05 ), which indicated no significant differences. Conclusion Target organisms can be isolated by the traditional method with a Ct value ≤ 25.5. Under the same broth Ct value, xylose lysine desoxycholate agar (XLD) and Hektoen enteric agar (HEA) are more preferable for isolation than R&F and MacConkey agar (MAC). R&F agar can be used for the preliminary determination of suspect organisms. More positive samples are confirmed by the test method than by the reference method, which suggested that the test method is better.
