摘  要:目的建立微波消解-氢化物发生原子荧光法测定紫薯中硒含量的方法。方法以微波消解对样品进行前处理,对微波消解参数、仪器条件、还原剂浓度、载流浓度、反应时间等试验条件进行了研究和优化;在最佳条件下,采用原子荧光法测定紫薯中硒的含量。结果硒标准曲线的回归方程为y=109.39x-7.31,此方法的检测限为0.22μg/L,在0.50μg/L^25.00μg/L浓度范围内线性关系良好,相关系数r=0.9999,相对标准偏差<5%。结论经试验证明,该方法应用于紫薯中硒的测定,硒含量在0.011 mg/kg^0.041 mg/kg之间,加标回收率为92.5%~102.5%,方法具有较高的灵敏度和准确度,操作简便而快速,可应用于紫薯中硒的测定,结果令人满意,可适合于基层实验室推广应用。Objective To establish the method for determination of selenium(Se) from purple potato by microwave digestion -hy- dride generation atomic fluorescence spectrometry. Methods To pre -treat samples with microwave digestion and investigate and op- timize the experimental conditions, such as microwave digestion parameters, instrument conditions, the concentration of the reducing agent, concentration carrier and reaction time. And at the best condition use atomic fluorescence spectrometry to check the determina- tion of selenium in purple potato. Results The regression equation of standard curve is for y = 109.39x - 7.31 selenium, under the optimum conditions, the detection limit of the proposed method was found to be 0.22 tJ-/L, the fluorescence intensity is linear to the concentration of Se in the range of 0.50 IZCe/L - 25.00 p.g/L, r = 0. 9999, the relative standard deviations were less than 5%. Conclu- sion Proved by the experiment, the proposed method was successfully applied to the determination of Se in purple potato, the con- tents were between in 0.011 mg/kg - 0. 041 mg/kg and the recoveries were in the range of 92. 5% - 102.5%, the method has a higher sensitivity and accuracy, the operation was simple and quick, it could be applied to the determination of Se in purple potato with sat- isfactory results and be suitable for the popularization and application of the simple laboratory.
