摘　　要：目的探讨口腔鳞状细胞癌(OSCC)中垂体瘤转化基因1(PTTG1)表达及其对上皮-间质转化(EMT)的作用。方法应用逆转录及定量PCR及蛋白印迹法检测58例OSCC组织及配对癌旁组织中PTTG1的表达;并采用RNAi技术下调口腔癌SCC9细胞系中PTTG1的表达,检测其对细胞中EMT相关基因E-cadherin表达的影响。结果PTTG1在OSCC组织与配对癌旁组织中的相对表达量分别为(3.046±1.137)和(0.975±0.434),差异有统计学意义(P<0.05)。OSSC组织中PTTG1的高表达与肿瘤临床分期及伴发淋巴结转移密切相关(P<0.05)。下调人口腔癌SCC9细胞系中的PTTG1的表达,细胞中E-cadherin的表达也减少,差异有统计学意义(P<0.05)。结论OSCC患者肿瘤组织中存在PTTG1的高表达,其高表达与肿瘤EMT存在相关性。Objective To investigate the expression of PTTG1 and its promoting effect for epithelial mesenchymal tran- sition （EMT） in oral squamous cell carcinoma, OSCC. Methods Quantitative RT-PCR （qRT-PCR） was carried out to detect the mRNA expression level of PTTG1 for 58 pairs of OSCC and normal control tissues. RNAi was used to knockdown PTYG1 expression in SCC9 cell line. E-cadherin level was detected before and after RNAi, using western blot. Resuits The relative expression of PTTGI in OSCC tissues （3.046± 1.137）was significantly higher than that of normal tissues （0.975±0.434） （P 〈 0.05）. Overexpression was closely related to the lymph node metastasis and clinical stage （P 〈 0.05）. E-cadherin level decreased remarkably after the knockdown of PTTG1 in SCC9 cell （P 〈 0.05）. Conclusion The overexpression of PTrGI in OSCC patients is closely related to OSCC clinical stage and lymph node metastasis, which promotes EMT progress.
