宫颈癌中JAK2/STAT3信号通道介导bax、bcl-2的表达

摘　　要：目的探讨JAK2/STAT3信号通道的激活及其介导bax/bcl-2在宫颈癌中的表达作用。方法选取本院2011-2014年宫颈癌患者40例,CINⅠ级组28例,CINⅡ/Ⅲ级组12例;选择正常宫颈15例为对照组。病理切片采用免疫组织化学方法测定P-JAK2、P-STAT3、bax、bcl-2的表达。结果1）对照组未见P-JAK2、PSTAT3蛋白表达;与对照组比较,CINⅠ级组及CINⅡ/Ⅲ级组表达增多（P〈0.05）;与CINⅠ级组比较,CINⅡ/Ⅲ级组表达增多（P〈0.05）。2）对照组可见少量bax、bcl-2蛋白表达;与对照组比较,CINⅠ级组及CINⅡ/Ⅲ级组表达增多（P〈0.05）;与CINⅠ级组比较,CINⅡ/Ⅲ级组的bax/bcl-2比率降低（P〈0.05）。3）CINⅠ级组和CINⅡ/Ⅲ级组通道蛋白P-JAK2、P-STAT3与bax/bcl-2比率呈正相关（P〈0.05）。结论宫颈癌患者JAK2/STAT3信号通道激活,可能与上调bax/bcl-2的表达和调节细胞凋亡有关。Objective To investigate JAK2/STAT3 signal pathway activation and the expressions of bax and bcl-2 in cervical carcinoma. Methods Forty cases of patients with cervical cancer in our hospital from 2011 to 2014 were analyzed,which of 28 cases were selected in the CIN Ⅰ and 12 cases were selected in the CIN Ⅱ/Ⅲ group. And then 15 cases of normal cervix were used as control group. Expressions of P-JAK2,P-STAT3,Bax, Bcl-2 were measured by immunohistochemical method and the data were analyzed by SPSS19. 0 software. Results 1 ） Expressions of P-JAK2 and P-STAT3 were not found in the control group. Compared with the control group, the expressions had increased more more in CIN I group and CIN Ⅱ/Ⅲ group （P〈0.05）. Then compared with CIN Ⅰ group,the expressions had increased in CIN Ⅱ/Ⅲ group （P〈0.05）. 2） The expressions of Bax and Bcl-2 protein in the control group was small. In contrast with the control group,the expressions had increased in CIN Ⅰ group and CIN Ⅱ/Ⅲ group （P〈0.05）. And compared with CIN I group, the Bax/Bcl-2 ratio had decreased in CIN Ⅱ/Ⅲ group （P〈0.05）. 3） The protein expressions of P-JAK2,P-STAT3 in CIN Ⅰ group and Bax/Bcl-2 ratio in CIN Ⅱ/Ⅲ group were positively correlated （P〈0.05）. Conclusion That the JAK2/STAT3 signaling pathway is activated in patients with cervical cancer may be associated with up-regulating expression of Bax/Bcl-2 in the mediation of cell apoptosis.
