摘 要:目的:研究正加速度(positive acceleration,+G z)暴露对实验性胃溃疡大鼠胃黏膜血流(gastric mucosal blood flow,GMBF)的影响,并探讨其影响溃疡愈合的相关机制.方法:选择♂SD大鼠40只随机分成4组,每组10只,分别标记为A、B、C、D组,A组为正常对照组,不做处理;对B、C、D组大鼠采用乙酸烧灼法建立胃溃疡模型,造模3 d后加速度处理大鼠,B组地面捆绑5 min(相当于+1 Gz恒速暴露),C组+5 Gz值旋转5 min,D组+10 Gz旋转5 min,隔日1次,共4次.1 wk后麻醉大鼠并固定,用MoorVMS-LDF2型激光多谱勒血流仪测定GMBF;将大鼠胃取出、展开,肉眼观察标本情况,用游标卡尺测量溃疡面积大小.结果:A组胃黏膜组织光滑、完整;B、C、D组大鼠胃窦小弯侧可见大小不等的溃疡病灶,且+Gz值越高,溃疡面积越大,C组(6.239mm2±0.597 mm2)与B组(1.602 mm2±0.490m m2)相比,溃疡面积明显增大(P<0.01);D组(9.511 mm2±1.857 mm2)溃疡面积比C组明显增大,差异有统计学意义(P<0.05).与A组GMBF(432.919 pu±11.598 pu)相比,B组GMBF(367.602 pu±10.133 pu)明显降低(P<0.01);加速度暴露后,C组GMBF(303.925pu±16.417 pu)、D组GMBF(246.750 pu±22.815 pu)显著低于B组(P<0.01);D组GMBF低于C组,差异有统计学意义(P<0.05).大鼠胃溃疡面积大小和GMBF量存在明显负相关关系(r=-0.911,P<0.05).结论:+Gz暴露导致实验性大鼠胃溃疡愈合延迟,溃疡愈合与GMBF密切相关,+Gz作用下GMBF下降可能是影响溃疡愈合的最直接的原因之一.AIM: To assess the effect of positive acceleration (+Gz) on gastric mucosal blood flow (GMBF) in rats with experimental gastric ulcer. METHODS: Forty male SD rats were random- ly and equally divided into four groups: A, B, C and D. Group A was normal rats without exposure to constant +Gz. Gastric ulcer was induced with acetic acid in rats of groups B, C and D. After 3 d, rats of groups B, C and D were exposed to constant +1 Gz, +5 Gz and+10 Gz for 5 min (once every other day, 4 times in all), respectively. After 1 wk, GMBF was measured with the MoorVMS-LDF2 laser Doppler blood flow meter, and the stomach was removed and observed macroscopically to measure the area of ulcer with a vernier caliper. RESULTS: Gastric mucosa tissue of rats of group A were intact. Different sized ulcer le- sions were observed in the gastric lesser curva- ture of rats of groups B-D. The higher the +Gz value, the larger the ulcer area. Compared with group B (1.602 mm2 ± 0.490 mm2), the ulcer area of rats of group C (6.239 mm2 ±0.597 mm2) was significantly larger (P 〈 0.01). The ulcer area of rats of group D (9.511 mm2 ± 1.857 mm2) was significantly bigger than that of rats of group C (P 〈 0.05). Compared with group A (432.919 pu ±11.598 pu), the GMBF of rats of group B (367.602 pu ±10.133 pu) was significantly lower (P 〈 0.01). After exposure to +Gz, GMBF of rats of groups C (303.925 pu ± 16.417 pu) and D (246.750 pu ±22.815 pu) was significantly lower than that of group B (P 〈 0.01). GMBF of rats of group D was significantly lower than that of group C (P 〈 0.05). There was a significant negative correlation be- tween gastric ulcer area and volume of GMBF (r = -0.911, P 〈 0.05). CONCLUSION: Exposure to +Gz delays gastric ulcer healing in rats, which is closely related to GMBF. The decrease of GMBF is probably one of the most direct factors affecting ulcer healing.
