乳腺癌中ID4基因启动子区甲基化状态及其表达

摘  要:目的检测乳腺癌中DNA结合抑制因子4(ID4)基因启动子区甲基化水平及其表达。方法采用焦磷酸测序法定量检测乳腺癌(A组,40例)及正常乳腺组织(B组,20例)标本中ID4基因启动子区甲基化水平,分析其与临床病理特征间的相关性;免疫组织化学SP法检测组织标本中ID4的表达。结果 A组ID4启动子区甲基化水平为(31.16±1.5)%,高于B组的(19.89±0.22)%(P<0.01);雌激素受体(ER)阳性乳腺肿瘤组织中ID4甲基化水平为(36.57±1.97)%,高于ER阴性组织的(27.91±1.83)%(P<0.01)。在乳腺癌组织和正常组织中,ID4的蛋白表达水平与其启动子甲基化水平呈负相关(r=-0.478,P<0.01)。结论 ID4基因在乳腺癌中呈高甲基化状态(特别在ER阳性的乳腺癌中);ID4基因启动子区甲基化可能是乳腺癌的发病机制之一。Objective To detect the promoter hypermethylation of inhibitor 4 of DNA binding (ID4) gene and its expression in breast cancer. Methods The methylation level of 1D4 promoter region was detected by pyrosequencing in breast tumor(group A, 40 cases) and normal tissue(group B,20 cases). The expression of ID4 was detected using immunohistochemistry method as well. The correlation between the rnethylation level of ID4 gene and clinical pathological characteristics was analyzed. The association between the expression and the methylation level was ananlyzed. Results The methylation level of ID4 promoter region was higher in group A than that in group BF(31. 16± 1.5) % vs. (19.89-1-0. 22)%] (P〈0. 01). The rnethylation level of ID4 gene was higher in estrogen receptor(ER) positive breast cancer tissues than that in ER negative [-(36.57±1.97)% vs. (27. 91± 1.83) %](P〈0. 01). The expression of ID4 protein was negatively correlated with the methylation level of ID4 in breast cancer and normal tissues (r=-0. 478, P〈0. 01). Conclusion Hyper- methylation of the promoter region of ID4 gene, especially in ER positive breast cancer, exists, which may be one of the mechanisms for breast cancer.
