摘　　要：目的:分析differentiated embryonic chondrocyte expressed gene 1(DEC1)在肺癌细胞系A549和BE1中对肿瘤增殖的调控作用。方法:应用siRNA和过表达质粒双向调控DEC1的表达后,通过MTT和克隆形成实验观察肿瘤细胞增殖的变化。并通过Real-time PCR和Western blot,在mRNA和蛋白水平,检测DEC1对细胞周期调控蛋白cyclinD1的调控作用。结果:在肺癌细胞系A549中,通过siRNA敲除内源性DEC1的表达后,cyclinD1在mRNA和蛋白水平的表达上调。同时,MTT和克隆形成实验的结果表明肿瘤的增殖能力显著增强;在肺癌细胞系BE1中,通过转入DEC1过表达质粒上调DEC1的表达后,cyclinD1的表达下调。同时,MTT和克隆形成实验的结果表明肿瘤的增殖能力显著降低。结论:在肺癌细胞系A549和BE1中,DEC1通过负向调控细胞周期蛋白cyclinD1,进而抑制肿瘤的增殖。Objective：To investigate the role of differentiated embryonic chondrocyte expressed gene 1 （DEC1）in regulating the proliferation of lung cancer cells. Methods： MTI＂ assay and colony formation were performed to examine the effect of DEC1 on the cell proliferation after dual regulation of DEC1. Meanwhile, Real - time PCR and Western blot were performed to investigate the role of DEC1 in regulation cyelinD1. Results：Downregulation of DEC1 by siRNA knockdown induced the up -regulation of cyclinD1 and promoted the growth and colony formation in theA549 lung cancer cell line,while overexpression of DEC1 downregulated the cyclinD1 and inhibited the growth and colony formation in the BE1 lung cancer cell line. Conclusion：In lung cancer cell line A549 and BE1, DEC1 inhibits the cell proliferation through the downregulation of cyclinD1.
