摘  要:目的:探讨一种在氧诱导视网膜病变(oxygen-induced retinopathy,OIR)动物模型中简单易行的视网膜铺片染色的技巧及新生血管定量的方法。方法:7 d龄SD大鼠建立OIR模型,分别于P12、P14、P16、P17、P24处死,做视网膜铺片,进行异凝集素染色,观察视网膜新生血管情况。结果:经过反复探索,视网膜可完整的铺展,经异凝集素染色后能清晰地呈现出视网膜新生血管情况。结论:掌握各步骤细节操作的技巧是保证大鼠视网膜铺片成功的关键。Objective:To explore an easy and effective technique for staining of retinal flat mounts and quatitative survey of neovessels in rat oxygen±induced retinopathy(OIR) model. Methods. Seven-day- old Sprague-Dawley rats were used to construct the OIR model and were sacrificed on P12, P14,P16, P17 and P24 respectively to stretch flat mounts which were then stained with fluorescein labeled GSL I -isolectin β to observe the retinal neovascularization. Results.Flat mounts,which were stretched completely, could show clearly retinal neovascularization after stained with fluorescein. Conclusion. It is critical in the preparation of flat mounts to master the skill of every step.
