摘  要:目的建立雄激素非依赖性前列腺原位癌动物模型,研究Pim-1基因及蛋白在该动物模型的表达。方法采用原位种植包埋法和外科手术去势技术,分别建立雄激素依赖、去势3d和雄激素非依赖性前列腺原位癌动物模型。分别采用基因芯片技术、酶联免疫法和免疫组织化学等实验方法,研究3组肿瘤组织中pim-1基因和蛋白表达的变化。结果 Affymetrix表达谱芯片技术检测结果显示,雄激素非依赖组Pim-1表达高于雄激素依赖组,差异有统计学意义,差异倍数为2.307 71;去势3d组与雄激素依赖组之间差异无统计学意义,差异倍数为1.108 67。ELISA检测结果显示,去势3d组血清睾酮浓度为(2.27±0.035)ng/mL,与雄激素依赖组的(9.02±0.99)ng/mL比较,差异有统计学意义,t=19.28,P<0.01;雄激素非依赖组为(0.29±0.068)ng/mL,与雄激素依赖组比较,差异有统计学意义,t=24.87,P<0.01;空白对照组为(9.23±0.78)ng/mL,与雄激素依赖组比较,差异无统计学意义,t=0.45,P=0.998。去势3d组PSA浓度为(0.17±0.032)ng/mL,与雄激素依赖组的(0.48±0.025)ng/mL比较,差异有统计学意义,t=21.82,P<0.05;雄激素非依赖组为(0.87±0.023)ng/mL,与雄激素依赖组比较,差异有统计学意义,t=31.53,P<0.05;空白对照组为0ng/mL,与雄激素依赖组比较,差异有统计学意义,t=41.80,P<0.01。免疫组织化学结果显示,雄激素非依赖组Pim-1蛋白表达量为0.024±0.001 9,明显高于雄激素依赖组的0.017±0.002 1,差异有统计学意义,t=8.27,P<0.05;去势3d组为0.018±0.001 3,与雄激素依赖组比较,差异无统计学意义,t=1.17,P=0.252。结论成功建立雄激素非依赖性前列腺原位癌动物模型,Pim-1与雄激素非依赖性前列腺癌有高度相关性。OBJECTIVE To investigate the different expression of Pim-1at gene and protein level in animal model from androgen dependent prostate cancer(ADPC)to androgen independent prostate cancer(AIPC).METHODS AIPC animal model was established by planting LNCaP tumor tissue and undergoing surgical castration.Affymetrix microarray technology and Immunohistochemistry and as well as ELLSA technology were carried out to compare the gene and protein expressions of Pim-1in three different groups of tumor tissue.RESULTS Affymetrix microarray results:the Pim-1expression in AIPC group had significantly differentiated 2.307 71 times higher than that in ADPC group;in castrated 3days group was 1.10867 times higher than that in ADPC group,and there was no significant differences in the Pim-1expression between castrated 3days group and ADPC group.ELISA result:The concentration of serum testosterone in castrated 3days group(2.27±0.035)ng/mL had significant difference with that in ADPC group(9.02±0.99)ng/mL,t=19.28,P〈0.01;and the difference had statistically significance between AIPC group(0.29±0.068)ng/mL and ADPC group,t=24.87,P〈0.01;but there was no difference in control group(9.23±0.78)ng/mL and ADPC group,t=0.45,P=0.998.The concentration of serum PSA in castrated 3days group(0.17±0.032)ng/mL had significant difference with that in ADPC group(0.48±0.025)ng/mL,t=21.82,P〈0.05;and the difference had statistically significance between AIPC group(0.87±0.023)ng/mL and ADPC group,t=31.53,P〈0.05;there was significant difference in control group 0ng/mL and ADPC group,t=41.80,P〈0.01.Immunohistochemical results:Using Image-Pro Plus(IPP)image analysis software on immunohistochemical image analysis,the expression of Pim-1protein level in AIPC group(0.024±0.0019)was higher than that in ADPC(0.017±0.002 1),and the difference had statistically significance(t=8.27,P〈0.05).The expression of Pim-1protein level between ADPC and castrated 3days group(0.018±0.001 3)had no si
