大蒜素对急性肺损伤大鼠Claudin-4调节作用研究

摘　　要：目的:探讨大蒜素对急性肺损伤大鼠的治疗作用及机制。方法:以雄性Wistar大鼠为对象通过颈动脉放血+气管LSP注射建立大鼠急性肺损伤模型,并通过静脉注射大蒜素予以治疗,以正常Wistar大鼠作为对照。术后通过病理检测、ELISA法等分析大蒜素疗效,并通过ELISA法、western blotting、PCR方法检测肺脏中Claudin-4表达变化。结果:经病理和ELISA检测显示大蒜素治疗组肺脏损伤情况明显好于急性肺损伤组,ELISA法、western blotting、PCR检测发现休克组大鼠肺脏Claudin-4表达较对照组明显降低,而以大蒜素治疗后Claudin-4表达可显著升高。结论:大蒜素可以减轻大鼠急性肺损伤,其机制可能是通过升高肺脏中Claudin-4的表达。Objective ： To investigate the effects of allicin treatment in rats with acute lung injury （ALI ）. Methods ： Male Wistar rats were as objects to establish acute lung injury （ALI）. Normal Wistar rats were used as control. We eval- uated the curative effect of allicin through pathological examination and ELISA and tested Claudin -4 expression in lung by ELISA, Western blotting and PCR. Results： After allicin treatment, pathological examination and ELISA test showed rats lung injury lighter compared with acute lung injury rats. ELISA, Western blotting and PCR results showed Claudin - 4 expression decreased in rats with acute lung injury and could increase significantly after allicin treatment. Conclusion： Allicin could alleviate acute lung injury. The mechanism might be due to up - regulating Claudin - 4 expression in lung.
