摘 要:建立一种高效液相色谱法测定花生和玉米中环匹阿尼酸含量的方法。样品经液液萃取净化处理,采用反相C18色谱柱Gemini C18(4.6 mm×250 mm,5μm),以乙腈:4%乙酸水溶液(三氟乙酸调pH值2.2)=55:45为流动相,流速1 mL/min,检测波长280 nm,对花生和玉米中的环匹阿尼酸进行检测。在50、100、200μg/kg添加量范围内,花生和玉米的平均回收率分别为83.9%~98.5%和86.3%~102.3%,方法的检出限为8μg/kg,线性范围为0.3~12.0μg/mL(R^2=0.9998)。所建立的方法快速、简便、准确,可以作为花生和玉米中环匹阿尼酸的检测方法。This study developed a reverse phase HPLC method for determing cyclopiazonic acid in peanut and corn. After samples were pretreated by liquid-liquid extraction, cyclopiazonic acid was separated on a Gemini C18(4.6 mm×250 mm, 5 μm) using a mobile phase composed of acetonitrile and 4% acetic acid(pH 2.2 adjusted with trifluoacetic acid)(55:45, v/v) at a flow rate of 1 mL/min and detected using a UVdetector set at 280 nm. At the spike levels of 50, 100, 200 μg/kg, the average recoveries for cyclopiazonic acid from peanut were between 83.9% and 98.5%, the average recoveries for cyclopiazonic acid from corn were between 86.3% and 102.3%. The limit of detection was 8 μg/kg for both peanut and corn, and the linear range was from 0.3~12 μg/mL(R^2=0.9998). The method was fast, easy and accurate. Therefore, the method developed can be used to determine cyclopiazonic acid in peanut and corn.
