摘 要:目的研究豹皮樟总黄酮(TFLC,Total Flavonoids of Litsea Coreana)对实验性2型糖尿病SD大鼠胰腺的保护作用及部分机制。方法以本实验室改良的方法建立2型糖尿病大鼠模型,将成模大鼠分为模型组和治疗组,治疗组分别采用小剂量TFLC(STFLC)和大剂量TFLC(LTFLC)治疗6周。取胰腺组织作胰岛素的免疫组化、氧化应激指标(GSH、T-SOD、Mn-SOD和T-AOC)和PTP1B的蛋白表达分析。结果 TFLC治疗组胰岛面积显著增加,胰腺的氧化应激明显减轻,胰腺中PTP1B的蛋白表达显著降低,且有一定的剂量效应。结论 TFLC具有明显的保护胰岛作用,其机制可能与减少胰腺中的氧化应激和PTP1B蛋白表达有关,且表现为一定的剂量效应。Objective To explore the effects and part mechanisms of TFLC on pancreasof experimental T2DM rat model. Methods The SD rat model of type 2 diabetes mellitus was constructed by improved method, then they were randomly allocated into model group and treatment group, rats in treatment group were treated with large and small dose of TFLC (LTFLC and STFLC) for 6 weeks respectively. The pancreatic tissues were used to analyze insulin content by using immunohistochemical method. The pancreas homogenates were used to detect the concentration of SOD, GSH, T - AOC and the protein expression of PTP1B respectively. Results The pancreatic islet areasin the treatment group were almost restored and the levels of SOD, GSH, T - AOC in pancreas were obviously decreased. The expression of PTP1B was significantly decreased in LTFLC group and showed definite dose effect. Conclusions TFLC has obvious protective effect on pancreas in T2DM rats, the mechanism may related with decreasing oxidative stress and PTP1B protein expression in pancreas, and showed definite dose effect.
