摘 要:长非编码RNA(lncRNA)是一类长度大于200nt不编码蛋白质的RNA,它们在生物体内发挥重要的调控作用。Non-polyA lncRNA是lncRNA的一个重要类型。为系统鉴定植物全基因组范围的non-polyA lncRNA,该文优化了拟南芥基因组范围鉴定新型non-polyA lncRNA的RNA样本纯化和测序方法,并且比较了3种不同的方法。结果表明:链特异性的non-polyA RNA长序列测序检测灵敏度更高,可以检测到更多低表达的lncRNA和更高比率的长序列lncRNA。因此,non-polyA RNA长序列测序适用于拟南芥全基因组范围的non-polyA lncRNA检测。该方法也可用于其它物种中non-polyA lncRNA的鉴定,为探究lncRNA功能提供帮助。Long non-coding RNAs(lncRNA)longer than 200 nt in length are implicated in most essential biological processes.Non-polyadenylated(non-polyA)lncRNAs contribute to a large class of lncRNAs.Non-polyA lncRNA on the genome-wide scale in plants were identified by optimizing the purification and sequencing methods to identify non-polyA lncRNAs in Arabidopsis thaliana.Three kinds of RNA sequencing methods were used with different in RNA samples and read lengths. The results show that strand-specific non-polyA RNA-seq with longer read lengths(101nt)may be the most sensitive and suitable assay method can detect lncRNAs with lower expression levels and higher ratio of long sequences.As a result, non-polyA RNA-seq with longer read lengths for genome-wide identification of non-polyA lncRNA in Arabidopsis thaliana.The method can be used to identify non-polyA lncRNAs in other species and can support studies of the function of lncRNAs.
