摘  要:目的考察胰蛋白酶抑制剂萘莫司他(FUT-175)对创伤性胰腺炎大鼠的治疗作用。方法采用压缩气流冲击法建立创伤性胰腺炎大鼠模型,以生理盐水和加贝酯分别作为阴性和阳性对照,考察FUT-175在造模后1 h腹腔注射给药后大鼠的生存率、血清淀粉酶和脂肪酶水平,以及胰腺组织病理学改变情况。结果与模型组相比,腹腔注射FUT-175后可显著提高创伤性胰腺炎大鼠生存率(60%vs.30%),降低血清淀粉酶[(3030.8±869.4)vs.(6126.5±791.3)U/L]和脂肪酶水平[(203.4±62.5)vs.(646.5±99.3)U/L],同时有效减轻胰腺组织损伤;与阳性对照药加贝酯相比,FUT-175给药后大鼠的生存率更高(60%vs.40%),血清淀粉酶[(3030.8±869.4)vs.(4111.4±683.1)U/L]和脂肪酶水平更低[(203.4±62.5)vs.(336.7±70.2)U/L]。结论胰蛋白酶抑制剂FUT-175对创伤引起的急性胰腺炎大鼠具有明显的治疗作用,值得进一步研究。Objective To study the curative effects of trypsin inhibitor nafamostat(FUT-175)on traumatic pancreatitis of rats. Methods Compressed air impact method was used to establish the rat models of traumatic pancreatitis and physiological saline and gabexate mesilate were used as the negative and positive control medicine in order to study the effects of intraperitoneal injection of FUT-175 in one hour after the modeling;it was observed the survival rate,serum amylase and lipase levels,and pathological changes of pancreatic tissue of rats. Results In comparison with the model group,after intraperitoneal injection of FUT-175,the survival rate of rats with traumatic pancreatitis(60% vs. 30%)was significantly improved,serum amylase[(3030. 8 ± 869. 4)vs.(6126. 5 ± 791. 3)U/L]and lipase levels ][( 203. 4 ± 62. 5 )vs.( 646. 5 ± 99. 3 )U/L ] were lowered,and pancreatic tissue injury was reduced effectively. In comparison with the positive control medicine gabexate mesilate,the survival rate of rats after administration of FUT-175 was much higher (60% Vs 40%)and serum amylase[(3030. 8 ± 869. 4)vs.(4111. 4 ± 683. 1)U/L]and lipase levels were much lower[(203. 4 ± 62. 5) vs.(336. 7 ± 70. 2)U/L]. Conclusion Trypsin inhibitor FUT-175 has obvious curative effects on acute traumatic pancreatitis of rats,and it is worthy of further study.
