摘 要:目的探讨EZH2蛋白和E-cadherin蛋白在乳腺癌组织中的表达及临床意义。方法采用免疫组织化学SP法检测62例乳腺癌组织中和20例乳腺增生组织中EZH2蛋白和E-cadherin蛋白表达,采用western-blot方法检测11例乳腺癌原发灶与转移淋巴结中EZH2蛋白和E-cadherin蛋白的表达,分析两者的表达情况与临床病例特征的关系以及两者之间的关联性。结果乳腺癌组织中EZH2蛋白表达高于乳腺增生组织(48.4%vs 10.0%,P<0.05);E-cad蛋白表达低于乳腺增生组织(51.6%vs 80.0%,P<0.05)。EZH2和E-cad蛋白表达均与乳腺癌临床分期、组织分级和淋巴结转移有关(P<0.05),但与肿瘤大小无关(P>0.05)。病期晚、分化差和易转移的肿瘤组织中,EZH2蛋白表达增加,E-cadherin蛋白表达降低。EZH2蛋白和E-cadherin蛋白表达呈负相关(r=-0.407,P<0.05)。EZH2蛋白在原发灶与转移淋巴结的表达无明显差异(P>0.05);E-cadherin蛋白在转移淋巴结的表达高于原发灶(P<0.05)。结论 EZH2蛋白和E-cadherin蛋白参与了乳腺癌的发生和发展,EZH2通过抑制E-cadherin表达促进肿瘤转移。[ Objective ] To detect the expression of EZH2 protein and E-cadherin protein in breast cancer tissue and their clinical significance. [ Method ] The SP immunohistoehemical analysis was used to detect the expression of EZH2 protein and E-eadherin protein in 62 breast cancer samples and 20 breast hyperplasia samples and the west- ern-blot was used to detect the expression of EZH2 protein and E-cadherin protein in eleven primary tumors and metastatic lymph nodes, the relationship of their expression with clinical pathological characteristics was analyzed and the correlation between the EZH2 protein and E-cadherin protein was understood. [ Result ] EZH2 protein ex- pression was higher in cancer tissue than in hyperplasia tissue (48.4% vs 10%, P 〈0.05); E-cadherin protein expression was lower in cancer tissue than in hyperplasia tissue(51.6% vs 80%, P 〈0.05). EZH2 protein expression and E-eadherin protein expression were significantly related to clinical stage, differentiation, and lymph node metastasis (P 〈0.05), but not to the tumor size (P 〉0.05). High expression of EZH2 protein and Low expression of E-cadherin protein were related the advanced stage, poor differentiation and lymph node metastasis in breast cancer patients. The expressions of EZH2 and E-eadherin in breast cancer tissues were inversely correlated (r =-0.407, P 〈0.05).There was no statistically significance in EZH2 protein expression between primary and metastatic foci(P 〉0.05) and E-cadherin protein expression increased in metastatic foci compared with metastatic foci (P 〈0.05). [ Conclusion ] EZH2 protein and E-cadherin protein are involved in initiation and progression of breast cancer which suggest that EZH2 may enhances breast cancer metastasis by inhibiting E-cadherin expression.
