摘 要:目的观察Ku70和特异性蛋白1(Sp1)在结肠癌组织中的表达变化,并探讨其意义。方法分别采用免疫组化SP法、RT-PCR法对结肠癌及对应癌旁正常组织中的Ku70和Sp1蛋白及mRNA进行检测,并分析二者间的相关性及与结肠癌临床病理参数的关系。结果结肠癌癌组织与癌旁正常组织中Ku70阳性表达率分别为68.9%(42/61)、36.1%(22/61),二者比较,P<0.05;Sp1阳性表达率分别为78.7%(48/61)、26.2%(16/61),二者比较,P<0.05。结肠癌和癌旁正常组织中Ku70 mRNA相对表达量分别为1.184±0.410、0.845±0.424,二者比较,P<0.05;Sp1 mRNA相对表达量分别为1.280±0.311、0.912±0.362,二者比较,P<0.05。Ku70 mRNA和Sp1mRNA在结肠癌组织中表达呈正相关(r=0.398,P=0.029)。Ku70和Sp1蛋白表达水平与结肠癌肿瘤大小、分化程度、淋巴结转移、TNM分期有关(P均<0.05)。结论 Ku70和Sp1在结肠癌中高表达,与结肠癌的侵袭、转移有关,二者可作为结肠癌发生、发展的预测指标。Objective To detect the expression of Ku70 and specificity protein 1 ( Sp1 ) in colorectal cancer tissue samples.Methods Immunohistochemistry and reverse transcription-polymerase chain reaction( RT-PCR) were used to de-tect the proteins and mRNA expression of the Ku70 and Sp1 in colorectal cancer samples and matched tumor-adjacent tissue samples respectively.The correlation beween the expression of Ku70 and Sp1 and clinical pathological parameter was also analyzed.Results The positive expression rate of Ku70 in tumor tissues and matched tumor-adjacent tissues was 68.9%(42/61) and 36.1%(22/61) respectively, the difference was statistically significant(P〈0.05).The positive expression rate of Sp1 was 78.6%(48/61) and 26.2%(16/61) respectively, the difference was statistically significant(P〈0.05). The relative expression of Ku70 mRNA in tumor tissues and matched tumor-adjacent tissues was 1.184 ±0.410 and 0.845 ±0.424 respectively, the difference was statistically significant(P〈0.05).The relative expression of Sp1 mRNA was 1.280 ±0.311 and 0.912 ±0.362, the difference was statistically significant(P〈0.05).The correlation of Ku70 mRNA and Sp1 mRNA was positive(r=0.398, P=0.029).The expression levels of Ku70 and Sp1 were associated with tumor size, tumor differentiation, lymph node metastasis, TMN stage(all P〈0.05).Conclusions The expressions of Ku70 and Sp1 in colorectal cancer are both higher than those in tumor-adjacent tissue, and are highly associated with the metastasis and infiltration in colorectal cancer.They may become new biomarkers in the occurrence and development of colon cancer .
