摘 要:目的观察CD47在链脲佐菌素(STZ)诱导的糖尿病(DM)大鼠模型肾脏中的表达情况,初步探讨CD47与DM肾损害的关系及其作用。方法将成年雄性SD大鼠80只随机分为正常对照组15只和DM组65只。采用STZ 50 mg/kg单次腹腔注射建立DM大鼠模型。监测各组大鼠空腹血糖(FBG)、尿蛋白(UP)、肌酐(Cr)、尿素氮(BUN);分别于2,4,8周处死各组大鼠,通过糖原染色(PAS)观察各组大鼠肾脏的病理改变,免疫组化观察CD47在各组大鼠肾脏的分布情况;分别对UP、Cr、BUN、肾脏病理损害、CD47蛋白表达进行相关性分析。结果 PAS染色可见DM组大鼠肾小球系膜基质增生,肾小球可见分叶现象等典型的糖尿病肾病损伤变化。与正常对照组相比,DM组大鼠的FBG、Cr、BUN、UP水平及CD47蛋白表达均明显增加,并且随着DM病程的延长而继续增加。DM组大鼠CD47的积分光密度与Cr、BUN、UP、肾小球硬化评分呈正相关。结论持续高血糖通过诱导DM大鼠肾小球CD47的高表达,引起细胞基质增生,参与DM大鼠肾脏的损伤和肾脏病变的进展。Objective It is to observe the expression of CD47 in the kidney of diabetes (DM) rat model induced by streptozotocin (STZ) and to discuss its role and the relationship with DM kidney injury. Methods A total of 80 male Sprague-Dawley rats were randomly divided into normal control group (n = 15) and DM group( n = 65). DM group were treated with an intraperitoneal injection dose (50 mg/kg) of STZ to establish the model of DM. The level of fasting blood glucose(FBG) , urine protein(UP), creatinine (Cr) and blood urinary protein (BUN) were monitored in each group;the rats in each group were sacrificed at 2, 4 and 8 weeks respectively to observe the pathological changes of kidney by periodic acid-schiff stain, the expression of CD47 in the rat kidney by immunohistochemical. The correlation between UP, Cr, BUN, renal pathological damage and CD47 protein expression were analyzed. Results Periodic acid-schiff stain showed mesangial and matrix proliferation in DM rats, some typical damage characteristics of diabetic nephropathy could be found in glomerular include leaf phenomenon, K - W nodules. Compared with normal control group, FBG, UP, creatinine, urea nitrogen and the expression of CD47 was significantly increased in DM rats(P 〈 O. 05 ) , and with the progress of DM continue to increase. The integrated optical density (IOD) of CD47 were positively correlated with Cr, BUN, UP and the grade of glomerular sclerosis. Conclusion Sustained hyperglyeemia can induce the expression of CD47 in glomerulus DM rat kidney damage and the progress of kidney disease.
