摘  要:目的:探讨Cyclin D1在结缔组织生长因子(CTGF)促进肺动脉平滑肌细胞(PASMC)增殖中的作用。方法:分离并培养SD大鼠肺动脉中膜平滑肌细胞,实验使用第3~7代细胞。经鉴定的PASMCs给予浓度分别为5 ng/ml、10 ng/ml、25 ng/ml、50 ng/ml、100 ng/ml CTGF培养48 h。采用WST-1检测细胞增殖;实时荧光RT-PCR和Western blot方法检测Cyclin D1m RNA及蛋白的表达。结果:随着CTGF浓度的增加,各组PASMCs的OD值均逐渐升高,当浓度达到5 ng/ml时与对照组比较,差异有统计学意义(P〈0.05),呈浓度依赖性;随着CTGF浓度的增加,各组PASMCs的Cyclin D1 m RNA和蛋白均较对照组表达升高。结论:CTGF通过上调Cyclin D1表达促进PASMC增殖。Objective:To investigate the role in Cyclin D1 in pulmonary artery smooth muscle cells(PASMCs)induced by connective tissue growth factor(CTGF). Methods: The PASMCs isolated from the pulmonary artery of SD rat were cultured in M199 culture system in vitro and the 3rd ~7th generation of PASMCs were used in the experiments. The cultured PASMCs were treated with 5 ng/ml, 10 ng/ml, 25 ng/ml, 50 ng/ml, 100 ng/ml CTGF,respectively. The proliferation of PASMCs was detected by WST-1 assay. The expression of Cyclin D1 was detected by real-time fluorescence RT-PCR and Western blotting. Results: The results of WST-1 test showed that, OD value in each group was gradually elevated with the increasing concentration of CTGF; when 5 ng/ml CTGF was used, the difference had statistical significance compared with the control group, which was concentration dependent. With the increase of CTGF concentration, both m RNA and protein expression of Cyclin D1 in each group were higher than that in the control group. Conclusions: CTGF may induce PASMC proliferation via up-regulated the expression of cyclin D1.
