摘  要:目的:检测分析内皮素-3(EDN-3)基因表达及其启动子甲基化在乳腺癌患者各种临床状态的变化。方法:采用实时定量PCR(RT-PCR)和甲基化PCR(MS-PCR)方法检测54例乳腺癌患者(实验组)及60例体检健康女性(对照组)外周血中EDN-3mRNA表达及其启动子甲基化发生率,比较以上两指标变化在乳腺癌患者不同临床状态的统计学差异。结果:实验组EDN-3mRNA表达较对照组显著降低,差异有统计学意义(t=-7.84,P<0.01),但这种降低在患者不同年龄、不同肿瘤组织类型和大小、有否区域淋巴结转移及远处转移之间差异均无统计学意义(P>0.05)。实验组EDN-3启动子甲基化检出率为55.56%(30/54),对照组未检出甲基化,差异有统计学意义(χ2=45.238,P<0.01);EDN-3启动子甲基化在上述不同临床状态时的差异亦无统计学意义(P>0.05)。结论:EDN-3mRNA转录下调和启动子甲基化可能与乳腺癌的发生有关,与患者临床病理状态无关。Objective:To investigate the mRNA expression and the incidence rate of methylation of endothelin-3 (EDN-3)promoter in patients with breast carcinoma in different clinical features.Method:The mRNA expreesi-on and methylation of EDN-3 gene in 54 patients with breast carcinoma (experimental group)and 60 healthy women (control group)were assessed with real-time quantitative polymerase chain reaction (RT-PCR)and methylation-spe-cific polymerase chain reaction (MS-PCR),respectively.The two index above between these gentic features and clinical features were analyzed.Results:The expression of EDN-3 mRNA was significantly lower in experimental group than that of control group (t=-7.84,P〈0.01).However there were no significantly differences in different clinical features including the age of patients,tumor tissue type and tumor size,lymph node metastasis and distant metastasis (P〉0.05).The detection rate of methylation of EDN-3 promoter was 55.56% (30/54)in experimental group,while the control group was not detected abnormal methylation samples,there were significantly differences between experimental group and control group(χ2=45.238,P〈0.01),but there were also no differences among all the clinical fea-tures (P〉0.05).Conclusion:The down-regulation of transcription of EDN-3 mRNA and the methylation of EDN-3 pro-moter might be related with the occurrence of breast cancer,but there were no differences among all the clinical features.
