摘  要:目的观察丹参酮ⅡA对缺血缺氧性脑损伤(HIBD)新生大鼠脑组织中白介素-1β(IL-1β)和肿瘤坏死因子(TNF-α)表达的影响。方法 7日龄新生SD大鼠60只,分层随机分为5组:假手术组(n=12),HIBD模型组(n=12),丹参酮治疗高剂量组、中剂量组、低剂量组,每组12只。模型组和丹参酮治疗组采用经典的RiceVannucci模型制备法,建立HIBD模型。于造模成功后及造模24 h,丹参酮治疗组给予丹参酮高、中、低剂量腹腔注射,模型组给予等量生理盐水腹腔注射。48 h后,取新生大鼠病变侧脑组织,以酶联免疫法检测脑组织中IL-1β和TNF-α的水平。结果 HIBD模型组及丹参酮高、中、低剂量治疗组脑组织中IL-1β和TNF-α的水平较假手术组显著升高,差异具有统计学意义(P<0.05),且丹参酮高、中、低剂量治疗组脑组织中IL-1β和TNF-α的水平较模型组显著降低,差异均具有统计学意义(P<0.05)。结论丹参酮ⅡA对新生大鼠脑缺血缺氧性脑损伤有一定的保护作用,其作用机制与下调脑组织中IL-1β和TNF-α的浓度有关。Objective To observe the effect of Tanshinone on expressions of interleukin-1β and TNF-α in neonatal rat with hypoxiaischemia brain damage. Methods HIBD model was established;75 SD neonatal rat aged 7 days were randomly divided into five groups:sham-operation group ,HIBD group and Tanshinone IIA high,medium ,low dose intervention group. After establishing the HIBD model, HIBD group were treated with intraabdominal injection of normal saline ,Tanshinone ⅡA high ,medium,low dose intervention group were treated with intraabdominal injection of Tanshinone ⅡA. Results In HIBD group and Tanshinone ⅡA intervention group, the expression levels of IL-1β, and TNF-α were increased significantly ,there were statistically significant difference compared with sham-operation group. And compared with HIBD group, the expression levels of IL-1β, and TNF-α of Tanshinone ⅡA were decreased significantly. Conclusion Tanshinone ⅡA could down-regulate the concentration of IL-1 β and TNF-α in brain tissue of newborn rats with hypoxic-ischemic encephalopathy. Besides, the decrement was positively associated with the Tanshinone ⅡA concentration.
