大肠癌FHIT和CHFR基因甲基化状态分析

摘  要:目的:分析大肠癌组织中FHIT和CHFR基因启动子区甲基化水平及其临床意义。方法:采用甲基化特异性聚合酶链反应(methylation specific PCR,MSP)技术检测46例大肠癌及癌旁正常组织的FHIT和CHFR基因启动子区甲基化状态。结果:大肠癌组织的FHIT和CHFR基因甲基化率分别为54.35%、49.15%,高于癌旁组织13.04%、8.69%,组间差异均有统计学意义(P<0.05),FHIT和CHFR基因甲基化状态与分化程度、淋巴结转移有显著相关性(P<0.05),与年龄、性别、肿瘤部位、Dukes分期无相关性(P>0.05)。结论:FHIT和CHFR基因启动子区的甲基化可能与大肠癌的发生、发展及预后相关。Objective:To analyze the methylation level of promoter region of fragile histidine triad ( FHIT) gene and checkpoint with FHA and ring finger ( CHFR) in colorectal cancer and its clini-cal significance.Methods:Methylation Specific PCR (MSP) was used to test methylation patterns of FHIT and CHFR gene in 46 colorectal cancer tissues and corresponding adjacent normal tissues . Results:The methylation rates of FHIT and CHFR were 54 .35%, 49 .15%respectively in color-ectal cancer, which were significantly higher than that in adjacent normal tissues ( 13.04%, 8 .69%) .The methylation status of FHIT and CHFR gene was correlated with differentiation and lymph node metastasis ( P〈0 .05 ) , but not associated with age , gender , tumor sites and Dukes Stage(P〉0.05).Conclusion: The methylation of promoter region of FHIT and CHFR may be correlated with occurrence , development and prognosis of colorectal cancer .
