摘 要:目的:探讨重组人BMP 7(bone morphogenetic protein-7,rh BMP 7)在体外对人小细胞肺癌(SCLC)细胞株SBC-3和SBC-5生物学行为的影响,为开发新的抗癌药物提供理论依据。方法:设置不同的rh BNP 7浓度梯度(0、50、100、150、200ng/ml)和时间梯度(24、48、72h)作用于SBC-3细胞和SBC-5细胞。MTT法检测细胞增殖情况,探索rh BMP 7的有效作用浓度和作用时间。选择有效作用浓度作用于SBC-3/5细胞,观察撤离rh BMP 7后细胞增殖的变化,并用流式细胞仪分析rh BMP 7对细胞凋亡和周期的影响。结果:rh BMP 7在体外对SBC-3细胞和SBC-5细胞的增殖有抑制作用,且有一定的浓度依赖性,有效作用时间为48h。rhBMP 7撤离后肿瘤细胞的增殖可以得以恢复。进一步研究还发现rh BMP 7可以抑制SBC-3和SBC-5细胞的迁移和侵袭能力。流式细胞仪分析rh BMP 7不影响SBC-5细胞的凋亡率,但会诱导SBC-3细胞的凋亡(P<0.05)。同时,rh BMP 7处理组中G1期的细胞比率有所升高,S期的细胞有所减少;并且这一影响在SBC-3细胞中更为明显。结论:rh BMP 7可以抑制SBC-3和SBC-5细胞的增殖,且抑制作用具有浓度和时间依赖性。rh BMP 7并不影响SBC-5细胞的凋亡,但会诱导SBC-3细胞的凋亡。因rh BMP 7可以增加G1期细胞的比率,推测可能是通过诱导细胞周期阻滞抑制肿瘤细胞增殖,从而为SCLC的治疗提供新的思路。Objective: To investigate the effects of biological behaviors induced by rh BMP 7 in small cell lung cancer( SCLC) cell lines SBC- 3 and SBC- 5. Methods: To figure out the effective concentration and functional acting time,human SCLC cells SBC- 3 and SBC- 5 were treated with rh BMP 7 at different concentration for 24,48 and72 h,the inhibition rates were determined by MTT assay. The optimal concentration was used to act on SBC- 3 and SBC- 5 cells,and then the change of proliferation after withdrawing rh BMP 7 was analyzed. In addition,transwell was applied to determine the invasive and migratory ability in the present of rh BMP 7. At last,cell cycle and apoptosis analysis were evaluated by flow cytometry. Results: In vitro,rh BMP 7 could inhibit the proliferation of SBC- 3and SBC- 5 cells at 100 ng / ml,which was in a dose- dependent and time- dependent manner. The SBC- 3 and SBC- 5 cells could restore their proliferative potency after withdrawing the BMP 7. After treatment for 48 h,the inhibition reached to the maximum. And rh BMP 7 could inhibit the invasive and migratory ability of SBC- 3 and SBC- 5 cells.The results of flow cytometry indicated that rh BMP 7 did not effect the apoptosis rates of SBC- 5 cell( P 0. 05),but can slightly increase the apoptosis rates of SBC- 3 cell. And the percentage G1 phase was higher when the rh BMP 7was added into the medium of SBC- 3 and SBC- 5 cells. Conclusion: rh BMP 7 inhibited the proliferation of SBC-3 and SBC- 5 cells depending on the concentration and exposing time. Meanwhile,rh BMP 7 could inhibit the invasion and migration. Although,rh BMP 7 did not affect the apoptosis of SBC- 5 cell,it could induce SBC- 3 cell to apoptosis. The cycle analysis revealed that rh BMP 7 could arrest SBC- 3 and SBC- 5 cells in G1 phase and inhibit the S phase entry.
