摘 要:目的研究核基质结合区结合蛋白质1(SATB1)基因及环氧化酶-2(COX-2)基因在胃癌组织中的表达及其意义。方法采用实时荧光定量聚合酶链反应检测30例胃癌组织及相应癌旁组织中SATB1基因、COX-2基因的表达,分析SATB1基因及COX-2基因表达与患者临床病理因素的相关性。结果胃癌组织中SATB1基因、COX-2基因表达量分别为癌旁组织的3.95倍和2.88倍,差异有统计学意义(P〈0.05);SATB1基因、COX-2基因在胃癌组织中的表达与淋巴结转移、肿瘤-淋巴结-转移分期、肿瘤浸润深度有关(P〈0.05),与性别、年龄、肿瘤分化程度无关(P〉0.05)。有淋巴结转移组SATB1基因、COX-2基因的表达量分别为无淋巴结转移组的1.99倍、2.38倍(P〈0.05);T4组SATB1基因、COX-2基因的表达量分别为T1~T2组的2.61倍、1.97倍(P〈0.05),Ⅰ期、Ⅱ期SATB1基因、COX-2基因的表达量差异无统计学意义(P〉0.05),Ⅲ~Ⅳ期SATB1基因、COX-2基因的表达量分别为Ⅰ期的2.99倍和2.52倍(P〈0.05)。胃癌组织中SATB1基因与COX-2基因的表达呈正相关(r=0.482,P〈0.05)。结论SATB1基因和COX-2基因参与了胃癌的发展、浸润及转移,二者具有协同作用。Objective To study the expression and significance of special AT-rich sequence-binding protein 1(SATB1) and cyclooxygenase-2(COX-2) in human gastric cancer. Methods The expression of SATB1 and COX-2 in thirty samples of gastric cancer tissues and cancer surrounding tissues were analyzed by real-time fluorescence quantitative polymerase chain reaction,meanwhile,the correlations between SATB1 gene,COX-2 gene expression and clinicopathological parameters were also analyzed. Results The expression of SATB1 and COX-2 in gastric cancer tissues was 3. 95 times and 2. 88 times respectively as much as those in cancer surrounding tissues(P〈0. 05),and this distinction had its statistical significance. Secondly,the expression of SATB1 and COX-2 in gastric cancer tissues had correlation with tumor node metastasis,tumor node metastasis stage and invasive depth(P〈0. 05),but was irrelevant to sex,age and tumor differentiation degree(P〈0. 05). Besides,the expression of SATB1 and COX-2 in node metastasis group was 1. 99 and 2. 38 times as much as that in the group without node metastasis(P〈0. 05). Group T4's expression was 2. 61 and 1. 97 times respectively as much as those of group T1- T2(P〈0.05). The expression distinction between stage Ⅰ and Ⅱ had no statistical significance(P〉0.05),however,the expression distinction between stage Ⅲ- Ⅳ and stage Ⅰ actually had its statistical significance;the expression of the former was 2. 99 and 2. 52 times respectively as much as the latter(P〈0. 05). The expression of SATB1 was positively correlated with the expression of COX-2 in gastric cancer tissues(r = 0. 482,P〈0. 05). Conclusion SATB1 and COX-2 are involved in the development,invasion and metastasis of gastric cancer,and the synergistic action is observed between the two.
