摘 要:目的探讨纳米炭-表阿霉素混悬液(carbon nanoparticles-epirubicin suspension,CNP-EPI)对人乳腺癌MDAMB-231细胞增殖和凋亡的影响。方法采用不同浓度的CNP-EPI处理MDA-MB-231细胞,CCK-8法测定不同浓度、不同时间对MDA-MB-231细胞增殖的抑制作用;根据测定的IC50选取5μg/m L CNP-EPI,相应浓度25μg/m L CNP及仅加培养基作用24 h后,观察细胞形态的变化,流式细胞仪(flow cytometry,FCM)测定细胞凋亡率。结果在1~200μg/m L浓度范围内,CNP-EPI对乳腺癌MDA-MB-231细胞增殖的抑制作用逐渐增强,48 h后仍有增加的趋势,但逐渐趋于稳定,且各时间段内对细胞的抑制率与药物浓度呈剂量依赖关系,与正常对照组比较,差异有统计学意义(P〈0.01),而相应各浓度CNP对细胞的增殖抑制作用不明显;5μg/m L CNP-EPI作用细胞24 h后,显微镜下可以看到大量的细胞死亡,正常对照组和相应浓度的CNP组细胞生长未见明显异常;流式细胞仪检测,CNP-EPI组作用24 h后,细胞的凋亡率为(14.37±6.25)%,死亡率可达(80.70±6.20)%,与相应浓度CNP组及正常对照组相比,差异有统计学意义(P〈0.05)。结论 CNP-EPI混悬液对人乳腺癌MDA-MB-231细胞增殖有明显抑制或杀伤作用,其抑制作用可能是通过其缓慢释放的表阿霉素直接杀伤癌细胞或诱导其凋亡实现。Objective To determine the effect of carbon nanoparticles-epirubicin suspension( CNPEPI) on the proliferation and apoptosis in human breast cancer MDA-MB-231 cells. Methods MDA-MB-231 cells were cultured with different concentrations of CNP-EPI in vitro. CCK-8 assay was used to determinate the inhibitory effect of CNP-EPI on the proliferation of MDA-MB-231 cells at different concentrations and for different times. According to the determination of IC50,5 μg /m L CNP-EPI( 25 μg /m L CNP) and only add culture medium were selected,and the cell morphology was observed with inverted microscopy,and cell apoptotic rates were determined by flow cytometry after 24 hours' treatment. Results CNP-EPI exerted gradually stronger inhibitory effect on the proliferation of MDA-MB-231 cells in a dose-dependent manner when the concentration ranged from 1 to 200 μg / m L for same time period,and the inhibitory effect was still in a increasing trend after48 hours' treatment,but slowly turned to stable. The inhibitory effect of CNP-EPI was significantly stronger in CNP-EPI treated cells than in normal control cells at all time points( P 0. 01). However,no such inhibitory effect was found in the MDA-MB-231 cells treated by different concentrations of CNP. For the cells treated by5 μg / m L CNP-EPI for 24 h,inverted microscopy observed that a great deal of cells were dead,but no obvious change of cell morphology was seen in the control cells and the cells treated by CNP. Flow cytometry indicated that the apoptotic rate was( 14. 37 ± 6. 25) % and the mortality reached to( 80. 70 ± 6. 20) % in the cells treated by 5 μg / m L CNP-EPI group for 24 h,which were significantly higher than those of CNP-treated cells and normal control cells( P 0. 05). Conclusion CNP-EPI exerts obviously inhibitory effect on the proliferation or killing effect in the human breast cancer MDA-MB-231 cells.
