细菌鞭毛蛋白衍生物CZLC331治疗小鼠溃疡性结肠炎的疗效

摘　　要：目的:研究细菌鞭毛蛋白衍生物CZLC331对小鼠溃疡性结肠炎(ulcerative colitis,UC)的治疗效果.方法:采用2,4,6-三硝基苯磺酸(2,4,6-trinitro benzenesulfonic acid,TNBS)溶液灌肠的方法建立小鼠UC模型,将24只实验小鼠分为模型组、治疗组和对照组,治疗组是针对模型小鼠造模后4 h腹腔注射3.2 mg/kg CZLC331蛋白.观察指标包括小鼠摄食水量、便血量、皮毛光泽度等一般状态、疾病活动指数(disease activity index,DAI),小鼠结肠组织黏膜和浆膜层变化及结肠黏膜损伤指数(colonic mucosa damage index,CMDI),小鼠结肠组织HE染色的病理改变.结果:与模型组相比,治疗组小鼠摄食(7 g/d±1.12 g/d)、摄水量(3 mL/d±0.78 mL/d)、活动性(14 min±5.32 min)明显增加,DAI评分(1.55±0.28)显著降低(P<0.05);治疗组小鼠结肠浆膜层充血、水肿,黏膜层出血及溃疡等现象亦有所减轻,CMDI评分(2.08±0.46)与模型组比较显著降低(P<0.05);治疗组结肠黏膜病理观察细胞排列结构比较整齐、少量杯状细胞消失及淋巴细胞和中性粒细胞浸.结论:腹腔注射细菌鞭毛蛋白CZLC331可以有效对抗结肠炎症对肠黏膜的损伤,对UC模型小鼠有明显的治疗作用.AIM： To assess the therapeutic effects of CZLC331 in mice with ulcerative colitis（UC）. METHODS： UC was induced in mice with 3,4,5-trinitrobenzenosulfonic acid（TNBS）. Twenty-four male Balb/c mice were randomly and equally divided into a control group, a UC model group, and a CZLC331 treatment group. After treatment, indexes including general status, disease activity index（DAI）, changes in the colon mucosa and muscularis layer, colonic mucosa damage index（CMDI）, and pathological changes of colon tissue were observed.RESULTS： Compared with the model group, food and water intake and activity increased significantly, and DAI score decreased significantly in the treatment group（P〈0.05）. Colonic serosal congestion, edema, mucosa bleeding and ulcers were significantly mitigated, and CMDI was significantly decreased in the treatment group compared with the model group（P〈0.05）. Pathological changes of colon tissue were also significantly reduced, and only disappearance of few goblet cells and lymphocyte and neutrophil infiltration were observed. CONCLUSION： CZLC331 has a significant therapeutic effect in UC mice.
