Optimization of Ultrasonic Extraction of Polysaccharide Extraction Technology from Stevia Rebaudiana by Response Surface Methodology

摘  要:以甜叶菊干叶为原料,采用超声法提取甜叶菊糖苷,探讨了提取次数、提取温度、超声时间和液料比对糖苷提取率的影响。通过单因素和响应面法确定最佳提取工艺条件为:提取次数2次,超声时间31 min,液料比30∶1,提取温度64℃,甜叶菊糖苷提取率可达到11.24%。The extraction procedures of polysaccharide from leaves of stevia rebaudiana were investigated with ultrasonic wave, according to the extraction times, extraction temperature, ultrasonic wave time and the ratio of liquid to material, respectively. The single factor extraction tests and response surface methodology were applied to determine the optimum technological condition. The results showed that the optimum extraction conditions of polysaccharides are as follows: the extraction time was 2, the ratio of liquid to material was 30∶1, extraction temperature was 64 ℃, ultrasonic wave time was 31 min. Under such conditions, the extraction rate is up to 11.24 %.
