周围神经损伤神经脊细胞caspase-3的表达与病理变化

摘　　要：目的探讨周围神经损伤神经脊细胞caspase-3的表达与病理变化。方法将90只SD大鼠随机分为3组:模型组30只制作大鼠周围神经损伤模型,假手术组30只坐骨神经显露后即关闭切口,空白对照组30只不做任何处理。采用免疫组织化学法测定各组制模后6h^5d神经脊细胞caspase-3蛋白的表达变化,HE染色及电镜观察神经脊细胞病理学变化,采用TUNEL法检测各组制模后6h^5d神经脊细胞的凋亡水平。结果免疫组织化学结果显示空白对照组及假手术组大鼠神经脊细胞caspase-3蛋白阳性表达、细胞凋亡较少,而模型组在周围神经损伤后6h即出现细胞凋亡,24~72h达高峰;caspase-3蛋白阳性表达也在6h明显增多,24h达高峰,72h表达减弱。神经脊细胞caspase-3蛋白的阳性表达与凋亡呈正相关(r=0.821,P<0.01)。结论周围神经损伤后神经脊细胞存在大量凋亡,其凋亡水平与caspase-3表达相关。Objective To investigate the changes in caspase-3 expression and histopathology in neural crest cells in peripheral nerve injury.Methods Ninety SD rats were randomly divided into three group,with 30 rats in each group.Peripheral nerve injury was induced in model group.The incision was closed after sciatic nerve exposure in sham-operation group.No treatments were per-formed in control group.The expression of caspase-3 in neural crest cells was detected by immu-nohistochemical staining,pathological change in neural crest cells by HE staining and electron mi-croscopy,and apoptosis of neural crest cells by TUNEL assay 6 hours-5 days after model estab-lishment.Results The expression of caspase-3 and apoptosis rate of neural crest cells were low in control group and sham-operation group.In model group,the apoptosis of neural crest cells were found at 6 hours after peripheral nerve injury and reached the peak between 24 and 72 hours after peripheral nerve injury.In addition,the expression of caspase-3 obviously increased at 6 hours, reached the peak at 24 hours and decreased at 72 hours.There was a positive correlation between caspase-3 expression and apoptosis in neural crest cells （r=0.821,P 〈0.01）.Conclusion Periph-eral nerve injury causes severe apoptosis in neural crest cells,and the apoptosis rate is correlated with caspase-3 expression.
