摘  要:目的为基层实验室提供一种简单有效的菌种保存方法。方法将常用菌种用甘油-生理盐水于-18℃低温冻存,每12个月进行1次活菌计数和特征鉴定。结果共成功保存18种菌,各菌种均能获得109/ml CFU以上的活菌。保存12个月时各菌种生物学特征与冻存前保持一致,保存24个月时,3种菌种的生物学特征有部分变化。结论 -18℃低温甘油-生理盐水保存菌种对常见菌种有效保存期可达12个月以上,并且简单易行,更适合条件有限的基层实验室使用。Objective To establish a simple and effective bacteria preserving method for basic level labs. Methods Common bacteria strains were cryo-preserved in glycerin-saline solution at-18 ℃, viable counting and feature identification were carried out every 12 months. Results Altogether 18 types of bacteria were successfully preserved, with a density of 10^9/ml CFU. The biological characteristics of the bacteria stayed the same as pre-freezing after 12 months and changed to some extent after 24 months and 36 months. Conclusion With features of simple and practicable, the method is suitable for preserving bacteria in 12 month for basic level labs.
