摘 要:目的:建立用高效液相色谱同时测定何首乌中已知有效成分二苯乙烯苷、大黄素、大黄酸、大黄酚、大黄素甲醚和芦荟大黄素含量的方法,并用此方法测定了12个市售地何首乌中有效成分的含量。方法:采用高效液相色谱法,Agilent SB-C18色谱柱(4.6mm×250mm,5μm),甲醇-0.1%磷酸溶液梯度洗脱,1.0m L/min,254nm,30℃。结果:6种化合物呈现较好的回归方程和相关系数,相关系数达到0.9997以上,二苯乙烯苷、大黄素和大黄素甲醚的平均回收率分别为100.94%、99.84%和100.27%。12个不同市售地何首乌中二苯乙烯苷和蒽醌类化合物的含量差别很大。结论:该测定方法快速准确,精密度高,稳定性好,大大缩短测定时间,对现行《中华人民共和国药典》方法有了很大的改进。市售药材测定结果为何首乌药材的质量控制和药用资源的开发利用提供了重要的参考。Objective: To establish a determination method for the contents of stilbene, emodin, rhein, chrysophanol, physcion and aloe-emodin in polygonum multiflorum by high performance liquid chromatography (HPLC). To detect the effective component of 12 commercial polygonum multiflorum by HPLC. Methods: HPLC was performed on a Agilent SB-C18 column (4.6mm×250mm, 5μm). Methanol-0.1% phosphoric acid water solution was used as gradient eluent at a flow rate of 1.0 mL/ min. The column temperature was set at 30℃, and UV detection wavelength was at 254nm. Results: The 6 chemical compounds mentioned above established a better regression equation and correlation coefficient. The correlation coefficient among 6 chemical compounds was up to 0.9997. The average recovery rat of stilbene, emodin and physcion was respectively 100.94%, 99.84% and 100.27%. There were significant differences in stilbene and anthraquinone compound contents of 12 commercial polygonum multiflorum. Conclusion: The advantages of HPLC method are fast and accurate, high precision, good stability and timesaving, which significantly improved the method in present pharmacopoeia. The determination result of 12 commercial polygonum multiflorum could provide an important reference for the quality control and the development of polygonum multiflorum resource.
