摘 要:评价广西苦丁茶不同生长期多糖含量的动态累积。在单因素考察基础上,通过响应面法优化得出最佳提取条件;采用紫外-可见分光光度法,以葡萄糖为对照品,于482 nm波长处,采用苯酚-硫酸法测定广西苦丁茶不同月份的多糖含量。多糖在5.83 mg/m L^38.54 mg/m L范围内呈良好线性关系(R2=0.997 9),平均加样回收率为98.618%,RSD为1.96%(n=5);样品的多糖含量在4月份、5月份最高,分别为96.6、82.4 mg/g,随后有所下降,12月份含量最低为47.7 mg/g。4月份苦丁茶多糖含量最高,如果以苦丁茶多糖生物活性作为标准,4月份是苦丁茶最佳采收期。To evaluate dynamic accumulation of total polysaccharides in different growth phase of Ilex Kudingcha C.J.Tseng from Guangxi. Based on the single factor, the response surface methodology was optimized to get the best conditions of extraction. UV-VIS spectrophotometry was used to detect the content of polysaccharide in 482 nm with glucose as reference substance. Anthrone-sulfuric acid method was used to determine the content of the total polysaccharides of Ilex kudingcha C. J. Tseng grown in different months. Polysaccharide in 5.83 mg/mL-38.54 mg/mL had a good linear relationship (R^2=0.997 9), the average sample recovery rate was 98.618%, RSD was 1.96%(n=5), the highest content of polysaccharide of samples grown in month is April (96.6 mg/g) and May (82.4 mg/g), then descend by with time when in December had the lowest amount just 47.7 mg/g. The highest content of polysaccharide of Ilex kudingcha C. J. Tseng is in April. Then using polysaccharide bioactivity as standard , April is the best harvest time of Ilex kudingcha C. J. Tseng.
