摘  要:目的初步建立前列腺腺癌基因组超甲基化谱,为前列腺腺癌特异性基因的超甲基化研究提供理论依据。方法本研究利用激光捕获显微切割技术和甲基化DNA免疫共沉淀芯片技术,初步建立3例前列腺腺癌及其癌旁腺体基因组启动子区甲基化谱,通过组间比较,筛选出一组前列腺腺癌基因组启动子区超甲基化谱。将筛选出的基因上传至Molecule Annotation System进行在线GO分析和Pathway分析。结果前列腺腺癌和癌旁腺体分别有7459个和7182个启动子区发生超甲基化,超甲基化基因占芯片总基因的33.1%和31.8%,其中65.6%的超甲基化启动子区为高Cp G密度启动子区。前列腺腺癌基因组超甲基化基因共273个。生物信息学分析提示启动子区超甲基化的基因涉及多个分子功能、生物过程和信号通路。结论前列腺腺癌和癌旁腺体之间存在大量启动子超甲基化改变,前列腺腺癌的发生发展可能是一个复杂的表观遗传学过程。Objective The initial establishment of prostate cancer genome hypermethylation was profiled to provide the evidence for the study of prostate cancer-specific gene hypermethylation. Methods Using laser capture microdisseetion and methylated DNA immunoprecipitation technique to initially establish the genomie methylation of the promoter region of the three cases of prostate adenoeareinoma and its paracaneerous glands. Screening a group of hypermethylation genes of prostate adenoear cinoma promoter region, Uploaded the genes to the molecule annotation system to perform gene ontology analysis and pathway analysis. Results Prostate adenocareinoma and adjacent gland had 7459 and 7182 methylation regions in the promoter, re- spectively. And the methylation incidence was 33.1% and 31.8%. A total of 273 hypermethylation genes in prostate adenocarei noma genome had been screened. The bioinformaties analysis suggested that these genes involved multiple molecular function, bi ological processes and pathways. Conclusion There are plenty of promoters hypermethylation changes between prostate adenocarcinoma and paraneoDlastic land. The development of prostate adenoeareinoma may be a comolex eoiuenetie processes.
