摘  要:目的利用荧光原位杂交技术检测拓扑异构酶(topoisomerase II,TOPII)在乳腺癌组织中的表达情况及相关临床意义。方法采用荧光原位杂交技术和免疫组织化学方法,对临床资料完整的264例浸润性乳腺癌标本分别进行TOPII基因及TOPII蛋白检测。结果 264例浸润性乳腺癌标本中,TOPII基因表达的阳性率为34%(91/264),且与乳腺癌临床分期、HER-2蛋白表达密切相关(P<0.05),而与患者年龄、淋巴结转移、肿瘤大小及ER、PR、TOPII蛋白表达(P>0.05)无关。结论拓扑异构酶II可能与乳腺癌的发生发展密切相关,在乳腺癌的治疗和预后中可能起重要作用。Objective To explore the expression of TOPII and its instruction in the treatment of the breast cancer. Methods The expression of TOPII was measured by fluorescence in situ hybridization (FISH) and immunohistochemical assay in 264 cases of infiltrative type of breast carcinoma tissues all of which had complete history date. Results The positive expression rate of TOPII gene was 34% in the breast carcinoma tissues, overexpression of TOPⅡ gene was significantly correlated with clinical stage and expression of HER-2 protein ( P 〈 0. 05 ), but it was insignificantly correlated with age, lymph node metastasis, tumor size and ER, PR and TOPII protein ( P 〉 0.05 ) . Conclusion The overexpression of TOPⅡ is closely corre lated with the occurrence and development of breast cancer and may play an important role in the evaluation of its curative effect and prognosis.
