摘 要:目的探讨香菇、辣椒、带鱼、羊肉和海虾5种食物对小鼠慢性变应性接触性皮炎(ACD)模型的影响。方法 36只小鼠用2,4-二硝基氟苯反复诱导建立慢性ACD模型后,随机分成5个处理组和1个对照组,分别予相应食物煎液和纯水灌胃。另设空白组。第4次激发后48h测量耳厚度和耳重量,计数炎症细胞,检测耳组织中IL-4和IFN-γmRNA水平。结果与空白组相比,造模后各组耳厚度差和IL-4mRNA表达水平明显升高,差异均有统计学意义(P均<0.05)。与对照组相比,辣椒和羊肉均能明显增加耳厚度差、耳重量差和炎症细胞数量(P<0.05),香菇、带鱼和海虾差异无统计学意义(P>0.05)。辣椒和海虾分别对耳组织中IL-4 mRNA和IFN-γmRNA表达水平有明显上调作用(P<0.01),而香菇、带鱼和羊肉差异无统计学意义(P>0.05)。结论 5种食物中,辣椒和羊肉可能通过不同机制加重小鼠慢性ACD模型的炎症反应。Objective To investigate the impact of five dietary foods ( mushroom, chili pepper, ribbonfish, mutton and sea shrimp) on chronic allergic contact dermatitis (ACD) in mice. Methods ACD mouse models were induced by repeated treatment of 2,4-dinitrofluorobenzene (DNFB) on thirty-six BALB/c mice. The ACD mice were randomly divided into 6 groups, respectively treated by each kind of food decoction (2g/mL)and pure water before the fouth elieitation. Six healthy mice were set as blank control. At 48 h after the fourth elicitation, the ear thickness, weight, the number of infiltrating inflammatory cells and the level of interleukin 4 (IL- 4) and interferonγ(IFN-γ) mRNA in the ear tissue were measured. Results Chronic ACD models were successfully induced by DNFB with a marked increase in ear thickness difference and IL-4 mRNA level compared with blank control. Compared with control group, the ear thickness, weight differences and the number of infiltrating inflammatory cells were markedly increased by chili pepper and mutton ( P 〈 0.05 ) , but not by mushroom, ribbonfish and sea shrimp ( P 〉 0.05 ) ; The level of IL-4 and IFN-γ mRNA in the ear were significantly increased respectively by chili pepper and sea shrimp (P 〈 0.01 ) , but not by mushroom, ribbonfish and mutton ( P 〉 0.05 ). Conclusion Among the five kinds of foods, chili pepper and mutton can aggravate the inflammatory reaction of chronic ACD model in mice possibly through different mechanisms.
