摘 要:目的:活化的小胶质细胞介导的神经炎症一直被认为是帕金森病(PD)的重要发病因素,而诱导型一氧化氮合成酶(i NOS)是一种重要的炎性蛋白,本文旨在探讨i NOS在活化小胶质细胞中的表达变化。方法:进行大鼠小胶质细胞原代培养,分为对照组及实验组,其中实验组加入脂多糖(LPS)1.0μg/ml分别刺激12 h和24 h。应用免疫荧光染色检测受刺激后小胶质细胞内i NOS定位分布。应用免疫蛋白印迹法观察受刺激后i NOS在小胶质细胞内水平变化。结果:实验组小胶质细胞内i NOS表达在12 h明显升高(P<0.01),24 h后表达恢复到对照组水平,染色显示i NOS主要表达于细胞质内。结论:在LPS诱导炎症模型中,i NOS在活化小胶质细胞内表达先升高而后恢复到原有水平,提示存在炎症反应的动态变化。Objective:Neuroinflammation mediated by activated microglial cells has long been regarded as one risk factor in Parkinson's disease( PD),and inducible nitric oxide synthase( i NOS) is an important inflammatory factor.This study was designed to test expression change of i NOS in microglial cells.Methods:Primary microglial cells of rats were harvest and divided into control and experimental groups.Experimental group was stimulated by LPS for 12 h and 24 h.Expression and location of i NOS in microglial cells were tested by immunofluorescence,and change of i NOS in microglial cells was tested by Western Blot.Results:Expression of i NOS in microglial cells increased at 12 h and went down to control level at 24 h after LPS( P〈0.01).i NOS was mainly located in cytoplasm of microglial cells.Conclusion:In LPS model,expression of i NOS in microglial cells increased firstly and then recovered.It suggested dynamic change of LPS-induced inflammatory respouse.
