摘  要:目的:研究铈离子对体外培养的骨髓间充质干细胞(BMSCs)成骨分化能力的影响。方法:分离培养小型猪骨髓间充质干细胞,分别用含有1×10-5mol/L CeCl3的成骨诱导液和单纯成骨诱导液进行培养;然后用碱性磷酸酶(ALP)试剂盒检测两组细胞诱导培养1、7、14、21 d的ALP活性;用实时荧光定量PCR分析两组细胞诱导培养14 d时的成骨相关基因Runx2、BSP、OCN mRNA的表达水平。结果:诱导培养1、7、14、21 d后,CeCl3组各时间点的ALP活性均较对照组显著增加(P<0.05);诱导14 d后,CeCl3组的各成骨相关基因Runx2、BSP、OCN mRNA表达水平均显著高于对照组(P<0.05)。结论:CeCl3能促进BMSCs的成骨分化。AIM: To investigate the effects of CeCl3 on the osteogenic differentiation of bone marrow mesenchymal stem cells( BMSCs). METHODS: BMSCs were obtained from porcine bone marrow aspiration and cultured inα-MEM medium with 10% FBS. The cells of third passage were exposed to inducing medium with or without CeCl3.The alkaline phosphatase( ALP) activity was measured. The gene expression of bone-related protein Runx2,BSP and OCN was analyzed by real- time RT-PCR. RESULTS: In the CeCl3 treated cells ALP activity,the gene expression level of bone-related marker Runx2,BSP and OCN were obviously higher than those in the control cells. CONCLUSION: CeCl3 may stimulate the osteogenic differentiation of BMSCs.
