摘 要:目的观察乏氧对中性粒细胞体外穿越牙龈上皮模型的影响。方法体外培养人牙龈上皮细胞并构建牙龈上皮模型,自外周血分离纯化中性粒细胞,使其在趋化剂的诱导下穿越牙龈上皮模型,以血平板计数和中性粒细胞髓过氧化物酶活性测定等方法检测中性粒细胞的穿越效率。结果’常氧环境下(21%O2),3h内66.8%-78.1%的中性粒细胞完成穿越牙龈上皮模型,但乏氧环境下(1%O2)穿越效率仅23.1%-27.5%。结论体外条件下乏氧抑制中性粒细胞穿越牙龈上皮模型。Objective To observe the effect of hypoxia on neutrophil transmigration across gingival epithelium model. Methods Human gingival epithelial cells were cultured in vitro and used to construct a gingival epithelium model. Neutrophils were separated and purified from human peripheral blood, and induced with a potent chemotaxin to transmigrate across the gingi- val epithelial model. The proportion of neutrophils that finished the transmigration in a given time was calculated with a hemacy- tometer, or by measuring the enzyme activity of neutrophil myeloperoxidase. Results In normoxia, 66.8%-78.1% of neutrophils finished the transepithelial migration in 3 hours, compared to only 23.1%-27.5% in hypoxia (1% oxygen). Conclusion Hypoxi- a inhibits neutrophil transmigration across gingival epithelium model in vitro.
