摘　　要：目的探讨肿瘤干细胞标记物八聚体结合转录因子4(Oct4)和乙酰脱氢酶家族成员A1(ALDH1A1)在乳腺癌组织中的表达及其临床意义。方法采用免疫组织化学方法检测208例乳腺癌组织和80例乳腺腺瘤组织中的Oct4和ALDH1A1蛋白的表达,分析其表达与临床病理参数及预后的相关性,并分析二者表达的相关性;同时采用western blot法检测20例乳腺癌组织和20例乳腺腺瘤组织中Oct4和ALDH1A1蛋白的表达,并分析二者表达的相关性。结果Oct4和ALDH1A1蛋白在乳腺癌组织中的阳性表达明显高于乳腺腺瘤组织(P<0.05)。Oct4和ALDH1A1的表达与乳腺癌组织学分级、组织学类型、TNM分期、淋巴结转移及三阴性乳腺癌密切相关。Oct4和ALDH1A1高表达的患者预后明显差于低表达的患者(P<0.05)。乳腺癌组织中OCT4和ALDH1A1表达具有显著相关性(P<0.05)。结论乳腺癌组织中,Oct4和ALDH1A1表达与组织学分级、组织学类型、TNM分期、淋巴结转移及三阴性乳腺癌密切相关;Oct4和ALDH1A1高表达提示乳腺癌预后不良;OCT4和ALDH1A1表达具有相关性。Objective To investigate the expression of ALDH1A1 and Oct4 in breast cancer and its clinical significance. Methods To detect the expression of Oct4 and ALDH1A1 in 208 eases of breast cancer and 80 cases of breast fibroadenoma by immunohistochemistry, analyze the relationship between the expression of OetT4 and ALDH1A1 and elinicopathological parameters and prognosis, and analyze the relationship between the expression of Oct4 and ALDH1AI. Meanwhile, to detect the expression of Oct4 and ALDH1A1 in 20 eases of breast cancer and 20 cases of breast fibroadenoma by western blot and analyze the relation- ship between the expression of Oct4 and ALDH1A1. Results The positive expression of Oct4 and ALDH1A1 protein in breast cancer tissue were significantly higher than that in the breast fibroadenoma. The expression of Oct4 and ALDH1A1 were closely related with histological grade, histological type, TNM stages, lymph node metastasis, and three negative breast cancer （P〈 0.05）. The prognosis of patients with high expression of Oct4 and ALDH1A1 was significantly worse than the patients with low expression （P〈0.05）. There was significant correlation between the expression of Oct4 and ALDH1A1 in breast cancer tissue （P〈 0.05）. Conclusions In breast cancer, the expression of Oct4 and ALDH1A1 were closely related with histological grade, histo- logical type, TNM stages, lymph node metastasis, and three negative breast cancer. High expression of Oct4 and ALDH1A1 pre- dicted the worse prognosis. There was significant correlation between the expression Oct4 and ALDH1A1 in breast cancer tissue.
