摘  要:目的建立短暂性脑缺血发作(TIA)小鼠模型,探讨灯盏生脉胶囊(DZSM)对其神经保护作用及可能的机制。方法采用尾静脉注射t-BHP及低氧方法成功制备了TIA样发作小鼠模型,分别给予DZSM 0.36 g/kg、DZSM 0.18 g/kg、DZSM 0.09 g/kg灌胃治疗,进行神经功能评分及行为学检测,化学比色法检测脑组织SOD、MDA、GSH-px的含量。结果高、中、低剂量DZSM均可明显改善TIA小鼠神经功能缺损评分情况,DZSM 0.36 g/kg、DZSM 0.18 g/kg可明显改善TIA小鼠卒中指数。TIA小鼠血清MDA明显增高、SOD、GSH-Px含量明显下降,高、中、低剂量DZSM可使TIA小鼠脑MDA含量均明显下降,而GSH-Px、SOD含量明显上升。结论灯盏生脉胶囊可降低TIA小鼠的神经功能缺损评分和卒中指数,其作用机制可能通过提高TIA小鼠体内抗氧化系统的活性,降低脂质过氧化反应,从而抑制t-BHP过氧化反应导致的神经、血管内皮细胞等氧化应激损伤来实现。Objective Toestablish mouse model of transient ischemic attack (TIA) and explore the neuroprotective roles and mechanism of Dengzhanshengmai (DZSM) capsule. Methods The TIA mice model were established successfully by injecting t-BHP and living in the hypoxia situation, and the intragastric injection with the doses of 0.36 g/kg, 0.18 g/kg, 0.09 g/kg DZSM was performed, then the Neurological Score and Ethology was detected, finally, the SOD, MDA, GSH-PX volume in cerebral tis- sues were detected. Results These three doses of DZSM could significantly decline the Neurological Score while the doses of 0.36 g/kg, 0.18 g/kg of DZSM could decline the Stroke Index of TIA mice. The volume of MDA was significantly increased meanwhile the SOD and GSH-Px were significantly decreased in the serum of TIA mice, however, these three doses of DZSM could decrease the volume of MDA and increase the SOD and GSH-Px respectively. Conclusion The DZSM capsule could decrease the Neuro- logical Score and Stroke Index of TIA mice, the mechanism of which may inhibit the t-BHP peroxide-induced oxidative injury in neurons and vascular endothelial ceils via increasing the activities of antioxidative system in TIA mice.
