摘 要:目的建立稳定表达pokemon乳腺癌细胞系,并观察pokemon对乳腺癌细胞系MCF-7侵袭转移能力的影响。方法构建表达pokemon慢病毒载体,将Pokemon表达慢病毒载体转染MCF-7细胞,通过Western blot方法检测稳定性表达pokemon MCF-7细胞中Pokemon-GFP融合蛋白的表达情况,应用Transwell试验及划痕实验观察pokemon的高表达对MCF-7细胞侵袭迁移能力的影响。结果成功建立稳定表达pokemon的MCF-7细胞系,pokemon可抑制MCF-7细胞侵袭能力。结论 Pokemon可抑制MCF-7细胞侵袭能力。Objective To establish MCF-7 cells with stablely ectopic pokemon expression and to assess the effect of pokemon on the invasion and migration of MCF-7 cells. Methods Immunofluorescence and Western blot were used to screen the MCF-7 cell clones with ectopic pokemon expression. Transwell and wound healing analysis were utilized to deter- mine the effect of pokemon on the invasion and migration of MCF-7 cells. Results The MCF-7 cell line with stablely ec- topic pokemon expression was established and Pokemon was demonstrated to suppress the invasion ability of MCF-7 cells. Conclusion Pokemon could suppress the invasion ability of MCF-7 cells.
