趋化因子受体CXCR2在结直肠癌组织中的表达及其临床意义

摘　　要：目的探讨结直肠癌患者组织中趋化因子受体CXCR2的表达水平及其对临床上结直肠癌侵袭、转移等预后的影响。方法以46例大肠癌、20例结肠息肉或腺瘤、30例正常结肠黏膜为研究对象,采用免疫组织化学SP法及RT-PCR法检测组织中CXCR2的蛋白质定量及m RNA半定量水平,并根据临床病理特征进行对照分析。结果CXCR2表达水平(蛋白水平及m RNA水平)在结直肠癌组织与大肠良性肿瘤组织中差异有统计学意义(P<0.05);CXCR2表达水平在肠癌组织与正常结肠组织中差异有统计学意义(P<0.05);CXCR2表达水平在肠癌淋巴结转移患者与未转移患者中差异有统计学意义(P<0.05);不同分化程度肠癌CXCR2表达水平差异有统计学意义(P<0.05);结直肠癌中CXCR2的表达与临床分期相关。结论结直肠癌细胞浸润、转移过程中可能有趋化因子受体CXCR2的参与,临床上对肠癌组织CXCR2的检测对其侵袭、转移生物学行为的预测具有重要的意义。Objective To investigate the significance of relationship between the expression of chemokine receptor CXCR2 in the tissue and invasion and metastasis in colorectal carcinomas. Methods The level of CXCR2 in the tissue were detected by immunohistochemical elivision plus method and RT-PCR method on protein and mRNA levels in 46 cases of colorectal carcinoma and 30 cases of benign colorectal tumor and 30 cases of normal colorectal tissue.The differ- ences were compared between benign and malignant diseases,clinical stages. Results The level of CXCR2 in tissue of colorectal carcinomas was obviously higher than that of benign tumor and normal colorectal （P〈0.05）.Compared with no lymph node metastasis,the CXCR2 level of lymph node metastasis was obviously higher （P〈0.05）.The intensity of CX- CR2 expression in tissue of colorectal carcinomas was correlated with tumor differentiation status （P〈0.05）.The CXCR2 level was positively related with clinical stages.And the levels of CXCR2 had ascending trends according to the clinical stages. Conclusion CXCR2 may play an important role in the invasion and metastasis of colon carcinoma.Monitoring the levels of CXCR2 in colon carcinoma tissue may have the important significance on forecasting the invasion and metastasis of colon carcinomas.
