骨桥蛋白及整合素β_3亚基与脑膜瘤恶性程度相关性

摘　　要：目的探讨骨桥蛋白、整合素β3亚基与脑膜瘤恶性程度的相关性。方法 2008年3月至2013年10月手术切除42例脑膜瘤,其中WHOⅠ级22例,Ⅱ级11例,Ⅲ级9例。另外,收集同期8例颅脑损伤减压术后正常脑组织标本作为对照。采用免疫组织化学染色分析检测各组骨桥蛋白和整合素β3亚基蛋白表达变化,采用逆转录酶聚合酶链反应方法检测各组骨桥蛋白和整合素β3亚基m RNA表达变化。结果正常脑组织未检测出骨桥蛋白与整合素β3亚基;随着脑膜瘤恶性程度的升高,骨桥蛋白和整合素β3亚基在脑膜瘤血管内皮及肿瘤细胞胞膜与胞浆中表达量显著升高(P<0.05)。结论骨桥蛋白和整合素β3亚基可能与脑膜瘤恶性程度相关。ObjectiveTo investigate the expressions of osteopontin（OPN） and integrin β3subunit in human meningiomas andtheir clinical meanings.MethodsThe expressions of OPN and integrin β3subunit in 8 samples of the human normal cerebral tissue and42 samples of human meningiomas, 22 were derived from grade Ⅰ menimgiomas, 11 from grade Ⅱ ones and 9 from grade Ⅲ ones, weredetected by immunohistochemical staining and semi-quantitative reverse transcription-polymerase chain reaction method.ResultsOPNand integrin β3subunit were not expressed in the human normal cerebral tissues. OPN and integrin β3subunit were expressed in the cellmembrane and cytoplasm and vascular endothelia of the menigiomas. The expressions level of OPN and integrin β3subunit weresignificantly higher in grade Ⅲ meningioma than those in grade Ⅱ menigioma（P〈0.05）, which were significantly higher than those ingrade Ⅰ menigioma（P〈0.05）. The expression of OPN was positively related to the expression of integrin β3subunit in the humanmeningiomas.ConclusionIt is suggested that the expression levels of OPN and integrin β3subunit are positively related to the malignantdegree of meningioma.
