摘 要:目的探讨血小板源性生长因子(PDGF)在食管鳞状细胞癌(ESCC)中的表达及其与肿瘤血管生成的关系。方法选择2011年10月至2012年1月新乡医学院第一附属医院胸外科行ESCC根治切除术的患者43例,取其手术切除的食管癌组织为ESCC组,同时选择其中25例ESCC切除标本上、下断端(距肿瘤边缘>5 cm)的正常食管上皮作为对照。采用免疫组织化学方法检测2组食管组织PDGF和微血管密度(MVD)-CD34的表达,并分析二者的相关性。结果 PDGF阳性产物主要在细胞质表达。ESCC组织中PDGF和MVD-CD34的表达水平均高于正常食管组织(P<0.05)。ESCC组织中PDGF的表达与MVD-CD34水平呈正相关(r=0.875,P<0.05)。结论血管生成在ESCC的发生、发展中有重要作用,PDGF可能在ESCC的血管生成中有促进作用。PDGF、MVD有可能成为ESCC抗血管靶向治疗的新方向。Objective To investigate the expression of platelet-derived endothelial growth factor(PDGF) in human esophageal squamous cell carcinoma(ESCC) and explore the relation between PDGF and angiogenesis in ESCC. Methods Forty-three cases of ESCC in the First Affiliated Hospital of Xinxiang Medical University from October 2011 to January 2012 were selected. The esophago diseased region of the patients which had been ablated in exairesis were collected as ESCC group. Normal esophago epithelium from the broken ends of excisional pecimen( the distance to tumor border 〉 5 cm) in 25 patients were collected as control group. The expression of PDGF and microvessel density( MVD)-CD34 in the two groups were detected by immunohistochemistry method and the correlation between them was analyzed. Results PDGF was mainly expressed in cytoplasm. The expression of PDGF and MVD-CD34in ESCC tissue was higher than that in normal esophageal epithelium. There was positive correlation between PDGF and MVD-CD34 in cancer tissues (r = 0. 875,P 〈 0.05 ). Conclusions Angiogenesis plays an important role in ESSC. PDGF may produce a promotion in angiogenesis of ESCC. PDGF and MVD are helpful for designing and finding effective anticancer drugs, providing new targets and initiatives for the management of ESCC.
