摘 要:目的探讨乳腺癌组织中PTEN、AKT的表达及与其临床病理及预后的相关性。方法采用免疫组化法,检测84例乳腺癌组织及30例乳腺腺瘤组织中PTEN、AKT蛋白的表达水平,分析其与乳腺癌临床病理特征的关系。结果与乳腺腺瘤组织比较,乳腺癌组织中PTEN蛋白阳性表达率显著降低(41.7%vs 73.3%,P<0.05)、AKT蛋白表达率显著升高(66.7%vs 20.0%,P<0.05)。PTEN阳性表达与乳腺癌肿瘤TNM临床分期、淋巴结转移相关。AKT阳性表达与乳腺癌TNM临床分期、肿瘤直径、淋巴结转移相关(P<0.05)。乳腺癌组织中PTEN表达与AKT蛋白表达呈线性负相关(γ=-0.752,P<0.05)。术后2年生存率PTEN蛋白阳性表达患者明显高于阴性表达患者,而AKT蛋白阳性表达患者明显低于阴性表达患者(P<0.05)。结论乳腺癌患者PTEN的表达缺失导致AKT的过度表达,PTEN、AKT蛋白表达的检测可能有助于预测乳腺癌的预后。Objective To investigate the expression of PTEN ,AKT in breast cancer ,and their relationship with clinical pathology and prognosis .Methods Expression of PTEN and AKT in 84 cases of breast cancer and 30 cases of breast fibroadeno-ma were detected by immunohistochemical staining ,and their relationship with clinicopathologic features were analyzed .Results Compared with breast fibroadenoma ,the positive rate of PTEN in breast cancer reduced significantly (41.7%vs 73.3%,P〈0.05),positive rate of AKT increased significantly (66.7%vs 20.0%,P〈0.05).The positive expression rate of PTEN was relat-ed with TNM clinical stage and lymph node metastasis (P〈0.05).The positive expression rate of AKT was related with TNM clinical stage,tumor diameter and lymph node metastasis (P〈0.05).PTEN and AKT was negatively correlated in breast cancer patients(γ=-0.752,P〈0.05).Postoperative 2-year survival rates of patients with positive expression of PTEN was significant-ly higher than those with negative expression ,and survival rates of patients with positive expression of AKT was significantly lower than those with negative expression (P〈0.05).Conclusion The under-expression of PTEN leads to the over-expression of AKT in patients with breast cancer ,detection of PTEN ,AKT protein expression may help to predict the prognosis of breast cancer .
