摘 要:通过简并引物克隆得到了桦褐孔菌及同属真菌共11个菌株的漆酶基因片段,并对其结构与同源性进行了分析。结果表明:11个目的扩增产物均含有2个内含子,且内含子位置保守,推导的氨基酸序列中均包含有保守的铜离子结合位点;DNA序列与氨基酸序列的相似度都颇高,基于2类序列构建的NJ系统进化树均能将除俄罗斯采集的桦褐孔菌菌株外归为1组,该漆酶基因片段可用于鉴定桦褐孔菌。Laccase gene fragments of 11 strains of Inonotus obliquus and several other species in the same genus were cloned by using degenerate primers, and its structure and homology were also ana?lyzed. The results indicated that two introns, the positions of which were highly conserved, were contained in all 11 PCR-amplified products, and conserved copper?binding sites ( His) were shown in the alignment of amino acid sequences. High similarities were reveled in both DNA and amino acid sequences, and the NJ trees constructed on the basis of the two sequences displayed that all I. obliquus strains expect Chaga were grouped together, suggesting this laccase gene fragment could be applied in species identification of I. obliquus.
